摘  要:目的研究纤维蛋白(fibrillarin,FBL)基因对大鼠早期胚胎发育的影响。方法选取健康、性成熟的大鼠30只,按照1:1的比例将雌鼠放置于雄鼠笼子里,同笼交配。采集大鼠胚胎,构建FBL上调、下调慢病毒载体,免疫荧光技术检测FBL在胚胎细胞中的表达,使用荧光定量RT-PCR技术检测FBL mRNA、Oct 4、Sox 2、FGF 4表达量,并作组间比较。结果 FBL在卵裂期胚胎中特异性染色不明显,而在囊胚中可被检测到有表达。上调组大鼠囊胚期FBL mRNA表达量、FBL表达量高于对照组,8-细胞阶段卵裂率、囊胚孵化率、Oct 4、Sox 2、FGF 4表达量低于对照组,差异有统计学意义(P<0.05);下调组大鼠囊胚期FBL mRNA表达量、FBL表达量低于对照组,8-细胞阶段卵裂率、囊胚孵化率、Oct 4、Sox 2、FGF 4表达量高于对照组,差异有统计学意义(P<0.05);下调组大鼠囊胚期FBL mRNA表达量、FBL表达量低于上调组,8-细胞阶段卵裂率、囊胚孵化率、Oct 4、Sox 2、FGF 4表达量高于上调组,差异有统计学意义(P<0.05)。结论下调FBL基因通过促进Oct 4、Sox 2的结合,调控FGF 4基因的表达,最终促进早期胚胎的分化。Objective To study the effect of fibrillarin(FBL) gene on early embryo development in rats. Methods Thirty healthy and sexually mature rats were selected and placed in male cages at a ratio of 1∶1 and mated with cages. The rat embryos were collected, FBL was up-regulated and down-regulated, and the expression of FBL protein in embryonic cells was detected by immunofluorescence technique. The expression of FBL mRNA, Oct 4, Sox 2 and FGF 4 was detected by fluorescence quantitative RT-PCR and compared among groups.Results The specific staining of FBL in cleavage stage embryos was not obvious, but it was detected in blastocysts. The expression of FBL mRNA and FBL protein in the blastocyst stage of the up-regulated group were higher than those in the control group. The cleavage rate, blastocyst hatching rate, Oct 4, Sox 2 and FGF 4 expression in the 8-cell stage were lower than those in the control group, the difference was statistically significant(P<0.05);the expression of FBL mRNA and FBL protein in the blastocyst stage of the down-regulated group were lower than that of the control group, and the cleavage rate, blastocyst hatching rate, Oct 4, Sox 2 and FGF 4 expression of the 8-cell stage were higher than that of the control group, the difference was statistically significant(P<0.05). The expression of FBL mRNA and FBL protein in the blastocyst stage of the down-regulated group was lower than that of the up-regulated group, 8-cell stage cleavage rate, blastocyst hatching rate, the expression of Oct 4, Sox 2 and FGF 4 was higher than that of the up-regulated group, and the difference was statistically significant(P<0.05). Conclusion Down-regulation of FBL gene regulates the expression of FGF 4 gene by promoting the binding of Oct 4 and Sox 2, and finally promotes the differentiation of early embryos.
