摘 要:目的:了解石家庄汉族献血人群血小板抗原HPA-18~21基因多态性。方法:采用PCR-SSP法对231例石家庄地区汉族无偿献血者进行HPA-18~21基因分型,并采用DNA测序验证。结果:231例石家庄汉族献血者中,全部样本均为HPA-18a/-18a、-19a/-19a、-20a/-20a纯合子,等位基因频率为100%;所有样本中227例为HPA-21a/-21a纯合子,4例为HPA-21a/-21b杂合子,并对杂合子进行了DNA测序证实,HPA-21a和HPA-21b的等位基因频率分别为99.13%和0.87%,与Hardy-Weinberg平衡吻合;在随机输血中HPA-21抗原不配合率为1.71%。结论:获得了石家庄汉族献血人群HPA-18~21基因多态性数据,为完善本地现有的HPA供者数据库、建立本地化已知HPA-18~21型别供者库提供了数据资料。Objective:To analysis the polymorphism of human platelet antigen HPA-18-21 gene in Han blood donors in Shijiazhuang.Method:Using SSP method to perform HPA-18-21 genotyping on 231 Han voluntary blood donors in Shijiazhuang,and verified by DNA sequencing.Result:All of the 231 Shijiazhuang Han blood donors were homozygous for HPA-18a/-18a,-19a/-19a,and-20a/-20a with an allele frequency of 100%.In all samples,227 were HPA-21a/-21a homozygotes and 4 were HPA-21a/-21b heterozygotes,then the heterozygotes were confirmed by DNA-sequencing,gene frequencies of HPA-21a and HPA-21b were 99.13%and 0.87%respectively,which fit Hardy-Weinberg equilibrium.The chance of incompatibility in random platelet transfusion was 1.71%for HPA-21.Conclusion:Gene frequencies of HPA-18-21 in Han blood donors in Shijiazhuang were obtained,which provided to improve the existing HPA database and establish a localized known HPA-18-21 donor database.
