益气化瘀补肾汤治疗腰椎间盘突出症的实验研究

摘　　要：目的研究益气化瘀补肾汤治疗腰椎间盘突出症的作用机制。方法①将大鼠分为正常对照组、模型组、阿司匹林组以及益气化瘀补肾汤低、中、高剂量组,复制了大鼠化学性神经根炎模型,模拟腰椎间盘突出症,在不同时间观察益气化瘀补肾汤低、中、高剂量组对化学性神经根炎模型大鼠炎性肉芽肿的形成及血液流变性的影响;②将小鼠分为正常对照组、阿司匹林组以及益气化瘀补肾汤低、中、高剂量组,观察益气化瘀补肾汤对小鼠耳廓肿胀和醋酸致小鼠腹腔毛细血管通透性的影响;③将小鼠分为正常对照组、模型组、阿司匹林组以及益气化瘀补肾汤低、中、高剂量组,观察益气化瘀补肾汤的镇痛作用和对血清中PGE2及c AMP浓度的影响。结果与模型组比较,中、高剂量的益气化瘀补肾汤可明显抑制化学性神经根炎模型大鼠炎性肉芽肿的形成,降低其全血粘度、血浆粘度、红细胞聚积指数(P <0.01),并能明显减少醋酸致小鼠扭体反应的次数,降低造模后升高的cAMP、 PGE2指数(P <0.01);与正常对照组比较,益气化瘀补肾汤减轻二甲苯致小鼠耳廓肿胀度和小鼠腹腔内炎性物质渗出(P <0.01)。结论益气化瘀补肾汤是通过抗炎、镇痛、改善血液流变性对腰椎间盘突出症起综合治疗作用的。Objective To study the action mechanism of Yiqi Huayu Bushen Decoction in the treatment of lumbar disc herniation.Methods ①Rats were divided into normal control group, model group, aspirin group as well as low, medium and high dose groups of Yiqi Huayu Bushen Decoction. The model of chemical radiculitis in rats was reproduced. Lumbar disc herniation was simulated. The formation of inflammatory granuloma and hemorheology of rats with chemical radiculitis in low, medium and high dose groups of Yiqi Huayu Bushen Decoction were observed at different time. ②Mice were divided into normal control group, aspirin group as well as low, medium and high dose groups of Yiqi Huayu Bushen Decoction to observe the influence of Yiqi Huayu Bushen Decoction on auricle swelling and abdominal capillary permeability induced by acetic acid. ③Mice were divided into normal control group, model group, aspirin group as well as low,medium and high dose groups of Yiqi Huayu Bushen Decoction to observe the analgesic effect of Yiqi Huayu Bushen Decoction and the influence on serum PGE2 and cAMP concentration. Results Compared with the model group, the medium and high dose groups of Yiqi Huayu Bushen Decoction could significantly inhibit the formation of inflammatory granuloma in rats with chemical radiculitis, reduce the whole blood viscosity, plasma viscosity and erythrocyte aggregation index(P <0.01), and significantly reduce the times of writhing reaction induced by acetic acid and the increase of cAMP and PGE2 concentration in mice(P <0.01). Compared with the normal control group, Yiqi Huayu Bushen Decoction could reduce the auricle swelling and inflammatory substance exudation in abdominal cavity induced by xylene of mice(P <0.01). Conclusions Yiqi Huayu Bushen Decoction has comprehensive effect on lumbar disc herniation by anti-inflammatory,analgesia and improving hemorheology.
