摘 要:以延胡索药材中5种生物碱的含量作为评价指标分析水煮、水蒸、醋煮和盐煮4种初加工方法的效果。采用甲醇加热回流法提取,采用高效液相色谱法检测含量。以乙腈-0.01%磷酸水溶液为流动相,按22∶78的比例进行等度洗脱,流速1.0 mL/min,色谱柱(Inertsil ODS-3)温度30℃,检测波长为283 nm。结果表明,原阿片碱、延胡索乙素、掌叶防己碱、小檗碱、脱氢紫堇碱5种生物碱线性范围分别为1.568~78.400、3.444~172.200、0.720~36.000、0.780~39.000、2.632~131.600μg/mL(r≥0.9993);平均回收率(n=6)为94.89%、97.13%、102.39%、98.06%、100.23%,相对标准偏差≤2.10%;4种延胡索加工方法中有效成分含量由高到低依次为水蒸>水煮>醋煮>盐煮,其中以蒸制5 min最佳。该方法简单、准确性高,可为延胡索产地初加工提供科学依据与技术支持。Taking the contents of five alkaloids in Corydalis yanhusuo as an evaluation index to analyze the effects of four origin processing methods:boiled,steaming,vinegar boiled and salt boiled.Five alkaloids were extracted by methanol heating and refluxing,and the contents were determined by high performance liquid chromatography.The acetonitrile-0.01% phosphoric acid aqueous solution was used as the mobile phase,and the isocratic elution was carried out at a ratio of 22∶78,the flow rate was 1.0 mL/min,the temperature of the column(Inertsil ODS-3)was 30℃,and the detection wavelength was 283 nm.The results showed that the linear ranges of five alkaloids,such as protopine,tetrahydropalmatine,palmetine,berberine and dehydrocortino,were 1.568-78.400,3.444-172.200,0.720-36.000,0.780-39.000,2.632-131.600μg/mL(r≥0.9993)respectively;the average recovery(n=6)was 94.89%,97.13%,102.39%,98.06%,100.23%,relative standard deviation≤2.10%;The active ingredients content from high to low was steaming>boiled>vinegar boiled>salt boiled,which was the best steamed for 5 min.The method is simple and accurate,and can provide scientific basis and technical support for the initial processing of the production area of Corydalis yanhusuo.
