摘 要:目前市场上白木香种苗主要为种子苗,由于缺乏系统选育,结香性状不稳定,因此,发展优良白木香嫁接无性系种苗具有重要的现实意义。‘热科2号’是一个前期筛选的优良白木香无性系,可以生产含较高色酮类化合物的沉香。为推广种植,开展扩大生产的研究。首先,采用基因组单碱基多态性(single nucleotide polymorphisms,SNPs)技术对‘热科2号’进行基因分型,验证群体遗传上的独立性及作为芽条培养的第一代嫁接苗和母树在遗传上的一致性。比较不同白木香品系作为砧木、不同条件下培养的砧木和芽条对芽片嫁接成活率的影响,同时,比较嫁接成活芽片在砧木锯顶后不同生长条件下的发芽率。SNP分型结果表明:‘热科2号’第一代嫁接苗与其母树在系统进化树上被聚集在同一分支上,有别于广东和其它海南白木香品系。嫁接试验结果表明:不同地域的白木香砧木对嫁接成活率无显著影响,但砧木和芽条的生长状况对嫁接成活率有显著影响,优化砧木和芽条的生长状况可提高芽片嫁接成活率至90.00%。嫁接成活芽片的发芽率对白木香嫁接整体成活率也有显著影响,在合适的条件下,芽片的发芽率可达80.00%,整体嫁接成活率达72.00%。该研究结果为‘热科2号’嫁接苗的生产提供了有效实用的方法。At present,Aquilaria sinensis planting materials on the market are mainly seedlings which are genetically various and stable in production of agarwood.It is of great practical significance to develop excellent planting materials of A.sinensis by grafting.A.sinensis’Reke2’is an excellent clone selected earlier that can produce agarwood with higher content of 2-(2-phenylethyl)chromone compounds.In order to promote expansion of planting,a method for expansion of A.sinensis’Reke2’was tested.A.sinensis’Reke2’were genotyped by using single nucleotide polymorphism(SNPs)to verify its population genetic independence and the genetic consistency of the first generation of graftings and their parent trees.Different clones or varieties used as root stocks,and rootstocks and budwood under different cultural conditions were tested to observe the effect of budpatches on the success of budgrafting and the sprouting rate of the budpatches under different conditions after the budded seedlings were cut back.The SNP genotyping indicated that the budded plants and their parent trees of A.sinensis’Reke2’were clustered on the same branch in the phylogenetic tree,which was different from that of other Hainan and Guangdong lines of A.sinensis.The grafting test showed that the survival rate of grafting was not significantly affected by the rootstocks of A.sinensis from different regions.However,the growth of rootstock and budwood significantly affected the survival rate of grafting.Under optimum growth conditions of rootstocks and budwood the budgrafting had a survival rate of upto 90.00%.The sprouting success of the grafted live buds significantly affected the success of budgrafting as well.Under normal conditions the sprouting rate of the buds of the budded plants was upto 85.00%and the survival rate of budgraftings was upto 75.50%.
