摘 要:目的:研究青蒿素对脂多糖(LPS)诱导肺泡上皮细胞炎性损伤的保护作用及其相关分子机制。方法:采用LPS(100 mg·L-1)刺激,建立肺腺癌上皮细胞系A549炎性损伤模型,给予青蒿素(20μM)预处理,研究青蒿素的抗肺细胞炎症效应。实时定量RT-PCR技术检测炎性信号RAI14蛋白及炎性因子IL-1β,IL-6和TNF-α的mRNA表达变化;免疫印迹分析脂多糖对RAI14蛋白水平的影响,并结合RAI14的小干扰RNA处理,揭示RAI14分子在LPS诱导肺炎症反应中的作用。结果:LPS诱导了肺上皮细胞RAI14的mRNA水平和蛋白含量的显著增加(P<0.05),且下调其表达能有效抑制LPS引起的炎症因子IL-1β,IL-6和TNF-α的mRNA水平升高(P<0.05)。给予青蒿素预处理,能显著下调LPS诱导的肺细胞RAI14的mRNA和蛋白水平升高(P<0.05),发挥抑制LPS诱导肺上皮细胞炎性反应的作用。结论:RAI14作为新识别的炎性信号分子,参与了LPS诱导的肺上皮细胞炎症反应。青蒿素能通过下调肺细胞RAI14基因表达,抑制LPS诱导的炎症反应。Objective: To study the protective effect of artemisinin on lipopolysaccharide(LPS)-induced inflammatoryinjury in alveolar epithelial cells and its potential molecular mechanism. Methods: LPS(100 mg·L-1)is used to establishan inflammatory injury model in lung adenocarcinoma epithelial cell line A549. Artemisinin(20 μM)is pretreated toinvestigate its anti-pulmonary inflammatory effects. RT-PCR is used to detect the mRNA levels of inflammatory signalRAI14 and inflammatory factors IL-1β, IL-6 and TNF-α. Immunoblot analysis is used to examine RAI14 proteinexpression, combined with RAI14 siRNA treatment to reveal the role of RAI14 in LPS-induced pulmonary inflammation.Results: LPS induces a significant increase in the mRNA and protein levels of RAI14 in lung epithelial cells(P<0.05).Down-regulation of RAI14 could effectively decrease the mRNA levels of inflammatory factors IL-1β, IL-6 and TNF-αstimulated by LPS(P<0.05). Pretreatment with artemisinin significantly inhibited RAI14 mRNA and protein expressionin lung cells induced by LPS(P<0.05), and attenuated the inflammatory response of lung epithelial cells induced byLPS. Conclusion: As a newly recognized inflammatory signaling molecule, RAI14 is involved in LPS-induced lungepithelial inflammatory response. Artemisinin inhibits LPS-induced inflammatory response by down-regulating RAI14 expression in lung epithelial cells.
