摘 要:目的为了更好地防控浙江省猪病毒性腹泻。方法从查明猪病毒性腹泻的病原流行情况入手,对2014—2016年度从浙江省部分猪场采集的小肠组织和粪便样本进行检测,根据PEDV(porcine epidemic diarrhea virus)高度保守的N基因核苷酸序列,设计合成了1对特异性荧光定量PCR(polymerase chain reaction)引物,并建立了一种可快速、灵敏、特异性检测PEDV的荧光定量PCR方法。PEDV检测用课题组建立的SYBR GreenⅠ实时荧光定量PCR检测方法,TGEV、RV检测用课题组建立的荧光定量RT-PCR检测方法。结果猪小肠病变组织中的PEDV的流行率66%,TGEV的流行率28%,RV的流行率7.3%;PEDV+TGEV混合感染的流行率23.3%;PEDV+TGEV+RV混合感染的流行率5.3%;在粪便样本中,PEDV的流行率26.7%,TGEV的流行率15%,RV的流行率4%,PEDV+TGEV混合感染的流行率10%,PEDV+TGEV+RV混合感染的流行率2%。结论目前引起猪场发生腹泻的主要病原为PEDV,不管是小肠组织还是粪便样本中都是PEDV流行比例最高,PEDV和TGEV混合感染比例较高,尽管PEDV+TGEV+RV共感染的比例较低,但是还是存在混合感染。因此,必须采取加强饲养管理、完善生物安全措施,实施疫苗免疫、中药喂服,对于疫情较严重猪场试行母猪病料返饲、控制继发感染及对症治疗等综合防治方案,才能达到较好的防治效果。Objective To prevent and control porcine viral diarrhea in Zhejiang province.Methods Based on the epidemiological investigation of porcine viral diarrhea,we tested small intestinal tissue and fecal samples collected from pig farms in Zhejiang Province during 2014 to 2016.We designed and synthesized a pair of specific fluorescence quantitative PCR primers and established a rapid,sensitive and specific fluorescence quantitative PCR method for detecting PEDV,according to the highly conserved N-gene nucleotide sequence of PEDV.The SYBR Green I real-time fluorescence quantitative PCR method established by our research group was used to detect PEDV,and the fluorescence quantitative RT-PCR method was also established to detect TGEV and RV.Results The prevalence rates of PEDV,TGEV,RV,mixed infection of PEDV+TGEV and PEDV+TGEV+RV in small intestinal lesions were 66%,28%,7.3%,23.3%and 5.3%,respectively.The prevalence rates of PEDV,TGEV,RV,PEDV+TGEV and PEDV+TGEV+RV in fecal samples were 26.7%,15%,4%,10%and 2%,respectively.Conclusion At present,the main pathogen causing diarrhea in pig farms is PEDV.The prevalence of PEDV is the highest in both small intestinal tissue and fecal samples.The co-infection rate of PEDV and TGEV is high.There is still a co-infection phenomenon of PEDV+TGEV+RV although its co-infection rate is low.Therefore,it is necessary to strengthen feeding management,improve biosafety measures,implement vaccination,feed traditional Chinese medicine,try out comprehensive prevention and control programs such as feeding pathological tissues of sows,controlling secondary infections and treating according to symptoms in piggery with serious epidemic situation so as to achieve better control effect.
