摘 要:为了研究载体菌株Δasd LH430(pYA3493)携带外源基因的能力,试验采用分子生物学方法构建了携带铜绿假单胞菌的保护性抗原基因F190-342-I21-83(F1I2)的重组表达质粒pYA-F1I2,将其重组到减毒的鼠伤寒沙门氏杆菌Δasd LH430(pYA3493)中,成功构建重组菌株Δasd LH430(pYAF1I2),并研究了重组菌株的遗传稳定性、表型、毒力。结果表明:构建的重组菌株Δasd LH430(pYAF1I2)能够稳定遗传所携带的F190-342和I21-83基因;重组菌株生长速度略低于亲本菌株,且遗传了亲本菌株的糖利用能力,在碳源利用方面没有改变;与亲本菌株相比,重组菌株毒力略有下降,对动物更加安全。说明重组菌株Δasd LH430(pYA-F1I2)可作为铜绿假单胞菌-沙门氏杆菌二联疫苗的候选菌株。To study the ability of carrying exogenous gene forΔasd LH430(pYA3493),a recombinant expression plasmid pYA-F1I2 carrying the protective antigen gene F190-342-I21-83(F1I2)of Pseudomonas aeruginosa was constructed.Then the plasmid was recombined intoΔasd LH430(pYA3493)with molecular biology methods,and the recombinant strainsΔasd LH430(pYA-F1I2)was successfully constructed.The genetic stability,phenotype and safety of recombinant strain carrying F1I2 gene were also studied.The results showed that F1 and I2 gene could be stably carried.The growth rate of the recombinant strain was slightly lower than the parent strain.Sugar fermentation test showed that the recombinant strain inherited the sugar utilization ability of the parent strain,and there was no change on using carbon source.The virulence of theΔasd LH430(pYA-F1I2)decreased slightly compared with the parent bacteria LH430.It showed that the recombinant strainsΔasd LH430(pYA-F1I2)were more safety for animals.It suggested that the recombinant strain ofΔasd LH430(pYA-F1I2)could be used as a candidate strain for the combined vaccine of Pseudomonas aeruginosa and Salmonella.
