摘 要:目的:探讨运动对雄性生长期小鼠骨内分泌FGF23——Klotho/FGFR1轴及相关因子表达影响,从骨骼调控角度为运动改善肾脏功能提供新的研究思路。方法:28只清洁级5周龄C57BL/6雄性小鼠随机分成对照组(C组)、跳跃组(J组)、游泳组(S组)和下坡跑组(R组),每组7只,进行50 min/天、6天/周、共8周运动训练,运动强度在65%~70%VO2 max。运动结束后,检测小鼠体重、单侧股骨湿重和胫腓骨湿重;Western blotting法检测骨中FGF23、肾脏中Klotho和FGFR1蛋白表达;RT-PCR法检测肾脏中NPT2a、NPT2c、Cyp24α1和Cyp27b1 mRNA表达;磷钼酸法检测血磷水平;酶联免疫血清检测1,25(OH)2D3水平。结果:(1)游泳组体重低于对照组和下坡跑组(P<0.05);跳跃组和下坡跑组股骨湿重高于对照组(P<0.05);跳跃组和下坡跑组股骨湿重高于游泳组(P<0.01);跳跃组和下坡跑组股骨湿重/体重水平高于对照组(P<0.01)。(2)跳跃组、游泳组和下坡跑组骨组织FGF23蛋白相对表达量高于对照组(P<0.05或P<0.01);下坡跑组肾脏FGFR1蛋白相对表达量高于对照组(P<0.05)。(3)下坡跑组NPT2a mRNA表达水平低于对照组(P<0.05);跳跃组和下坡跑组Cyp24α1 mRNA表达水平高于对照组(P<0.01),下坡跑组Cyp24α1 mRNA表达水平高于游泳组(P<0.01);下坡跑组Cyp27b1 mRNA表达水平低于对照组(P<0.05)。(4)游泳组和下坡跑组血磷水平低于对照组(P<0.01);跳跃组、游泳组和下坡跑组血清1,25(OH)2D3含量低于对照组(P<0.05或P<0.01)。结论:下坡跑运动显著提高骨骼中FGF23水平,并通过FGF23--Klotho/FGFR1轴,降低NPT2a和NPT2c表达,降低血磷水平;提高Cyp24α1表达,降低Cyp27b1表达,降低1,25(OH)2D3水平,其作用效果优于跳跃运动和游泳运动。Objective To explore the effect of exercise on the expression of Fibroblast Growth Factor 23(FGF23)—Klotho/Fibroblast Growth Factor Receptor 1(FGFR1)axis and related factors via the bone endocrine in the male mice,and to provide new research ideas for the improvement of kidney func tion from the perspective of bone regulation.Methods Twenty-eight 5-week-old C57BL/6 male mice were randomly divided into a control group(group C),a jumping group(group J),a swimming group(group S)and a downhill running group(group R),each of 7.The exercise was performed 50 min/d,6d/w for 8 weeks,and the exercise intensity was 65%~70%VO2 max.After the exercise,the body weight,femur wet weight and tibiofibula wet weight were measured.The Western blotting was used to detect FGF23 in bone,as well as Klotho and FGFR1 protein expression in the kidney.The reverse transcription-polymerase chain reaction was used to test the mRNA expression of Na+-dependent phos phate co-transporter 2a(NPT2a),Na+-dependent phosphate co-transporter 2c(NPT2c),Cyp24α1 and Cyp27b1.The serum phosphorus level was detected using the phosphomolybdic acid method,while the 1,25(OH)2D3 level was measured using the enzyme-linked immunosorbent serum.Results(1)Com pared with group C and R,the body weight decreased significantly in group S(P<0.05).Compared with group C,the weight of femur increased significantly in group J and R(P<0.05).Compared with group S,the weight of femur increased significantly in group J and group R(P<0.01).Compared with group C,the ratio of femur/weight increased significantly in group J and group R(P<0.01).(2)Com pared with group C,the protein expression of FGF23 increased significantly in group J,S and R(P<0.05 or P<0.01),while the protein expression of FGFR1 increased significantly in group R(P<0.05).(3)Compared with group C,the mRNA expression of NPT2a decreased significantly in group R(P<0.05),and the mRNA expression of Cyp24α1 increased significantly in group J and group R(P<0.01).Compared with group S,the mRNA expression of Cyp24α1 decre
