摘  要:目的:探讨白花败酱草对宫颈癌Hela细胞凋亡及对其可能的作用机制。方法:将不同浓度(0、0.1、0.2、0.4、0.8mg/ml)的白花败酱草提取物分别作用于宫颈癌Hela细胞,在不同时间点(24、48、72h)采用MTT法检测细胞的增殖情况;采用流式细胞仪检测Hela细胞的凋亡率及Bcl-2和Bax的表达情况。结果:与对照组相比,白花败酱草提取物对各组Hela细胞的增殖均有抑制作用;不同浓度的白花败酱草对Hela细胞生长均有抑制作用,与剂量、时间正相关。随白花败酱草提取物浓度降低,Bcl-2蛋白的表达降低,Bax蛋白的表达升高。结论:白花败酱草可促进宫颈癌Hela细胞的凋亡,其作用机制可能与提高Bax活性,降低Bcl-2活性有关。Objective:To invest the role of patrina villosa juss induced apoptosis of human cervical cancer Hela cells and its mechanism.Methods:Hela cells were exposed to the concentrations of Patrina villosa juss 0,0.1,0.2,0.4 and 0.8 mg/ml for 24,48,72h.Cell proliferation was detected by MTT assay.Apoptosis and expression of Bcl-2 and Bax protein was determined by double stained assay with Annexin-Ⅴand PI.Results:After Hela cells were cocμltured with 0,0.1,0.2,0.4,0.8 mg/ml patrina villosa juss for 24,48,72h in vitro,the cell proliferation were inhibited in time and dose dependent manner.When the Hela cells were cμlture for 48h by atrina villosa juss 0.2,0.4,0.8mg/ml,the total apoptosis rate was increased,which had ststistical significance compared with control group(P<0.05).The expression level of Bax increased after Hela cells were treated by Patrina villosa juss 0,0.2,0.4 mg/ml with a concentration effect relationship,while Bcl-2 is decreased(P<0.05).Conclusion:Patrina villosa juss can inhibit the proliferation and induce the apoptosis of Hela cells in vitro,one of its possible mechanisms is that Patrina villosa juss regμlate antiapoptotic and proapoptotic proteins.
