摘  要:目的:观察参蛤散对离体大鼠胸主动脉的舒张作用,并探讨其可能的作用机制。方法:运用离体组织灌流技术,记录参蛤散对苯肾上腺素(phenylephrine,Phe)预收缩的离体大鼠胸主动脉环的作用,并观察、分析左旋硝精氨酸甲酯(NG-nitro-L-arginine methyl ester,L-NAME)、环氧化酶抑制剂吲哚美辛(Indomethacin,Indo)、鸟苷酸环化酶剂{1H-[1,2,4]-oxadiazolo-[4,3-α]-quinoxalin-1-one,ODQ}、去血管内皮等不同干预条件对参蛤散舒张血管作用的影响。结果:参蛤散(10~200 g·L^-1)对Phe(1μmol·L^-1)预收缩的离体大鼠胸主动脉环有浓度依赖性的舒张作用。L-NAME(100μmol·L^-1)、Indo(1μmol·L^-1)、ODQ(1μmol·L^-1)均可显著抑制参蛤散对血管的舒张作用。在去内皮血管与经L-NAME+Indo预处理的血管环中相比,参蛤散对离体大鼠胸主动脉环的舒张作用无明显差异。结论:参蛤散对Phe预收缩的大鼠胸主动脉具有显著的舒张作用,并呈量效关系,其舒张血管作用呈现内皮依赖性,可能与内皮舒张因子一氧化氮和前列环素有关。Objective: To observe the diastolic effect of Shen Ge Powder on isolated thoracic aorta in rats and explore their possible mechanism. Methods: Isolated tissue perfusion technique was used to record effects of Shen Ge Powder on phenylephrine( Phe)precontracted thoracic aortic rings in rats. Effects of different intervention conditions such as NG-nitro-L-arginine methyl ester( LNAME),cyclooxygenase inhibitor Indomethacin( Indo),guanylate cyclase agent { 1 H-[1,2,4]-oxadiazolo-[4,3-alpha]-quinoxalin-1-one( ODQ) } and de-endothelium on vasodilation of Shen Ge Powder were observed and analyzed. Results: Shen Ge Powder( 10-200 g·L^-1) has a concentration-dependent relaxation effect on the isolated rats horacic aortic rings pre-contracted by Phe( 100 μmol·L^-1). L-NAME( 100 μmol·L^-1),Indo( 100 μmol·L^-1) and ODQ( 100 μmol·L^-1) could significantly inhibit the vasodilation effect of Shen Ge Powder on blood vessel. Compared endothelium-denuded vascular with the vascular rings pretreated with L-NAME + Indo,the vasodilation effect of Shen Ge Powder on isolated rat thoracic aortic rings was not significantly different. Conclusion: Shen Ge Powder has significant vasodilation effect on the precontracted rats thoracic aorta with Phe in a doseeffect relationship. Its vasodilation is endothelial dependent,which may be related to endothelial relaxing factors Nitric oxide( NO)and Prostacyclin( PGI2). Conclusion: Shen Ge Powder has significant vasodilation effect on the precontracted rats thoracic aorta with Phe in a dose-effect relationship. Its vasodilation is endothelial dependent,which may be related to endothelial relaxing factors Nitric oxide( NO) and Prostacyclin( PGI2).
