摘 要:目的:观察七味白术散加减对糖尿病模型大鼠氧化应激及肾功能的干预作用。方法:SD大鼠经高脂饲料喂养2周后,腹腔注射链脲佐菌素,建立类似于气阴两虚型糖尿病模型大鼠。模型大鼠随机分为模型组,二甲双胍组(0.2 g·kg^-1),七味白术散高剂量组(40.4 g·kg^-1)、中剂量组(26.9 g·kg^-1)、低剂量组(13.5 g·kg^-1),每组各15只。模型组、空白组大鼠给予等量的纯净水。每天灌胃给药1次,连续给药8周。观察大鼠一般状况,检测空腹血糖、血清超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)水平、谷胱甘肽过氧化物酶(glutathion peroxidase,GSH-Px)、血清肌酐、尿素氮等水平。结果:模型组血糖水平均显著高于空白组(P<0.05),糖尿病模型造模成功。与模型组比较,二甲双胍组,七味白术散高剂量组、中剂量组空腹血糖显著降低(P<0.05)。与模型组比较,七味白术散高剂量组、中剂量组SOD活性、GSH-Px活性显著升高,MDA、血清肌酐、尿素氮水平显著降低(P<0.05);七味白术散低剂量组SOD活性显著升高(P<0.05),MDA、尿素氮水平显著降低(P<0.05)。结论:七味白术散加减可降低气阴两虚型糖尿病模型大鼠空腹血糖水平,抑制氧化应激反应,改善肾功能。Objective: To observe the effect of Qiwei Baizhu powder on oxidative stress and renal function in diabetic rats. Methods:SD rats were fed with high-fat diet for 2 weeks,then streptozotocin was injected intraperitoneally to establish a diabetes model similar to the deficiency of both qi and yin. Model rats were randomly divided into model group,metformin group( 0. 2 g·kg^-1),Qiwei Baizhu powder high dose group( 40. 4 g·kg^-1),medium dose group( 26. 9 g·kg^-1),low dose group( 13. 5 g·kg^-1),15 rats in each group. Rats in model group and blank group were given equal amount of pure water. The drug was given by gavage once a day for 8 weeks. The levels of fasting blood glucose,superoxide dismutase( SOD),malondialdehyde( MDA),glutathione peroxidase( GSH-PX),serum creatinine and urea nitrogen were measured. Results: The blood glucose level in the model group was significantly higher than that in the blank group( P < 0. 05). Compared with the model group,the fasting blood glucose of metformin group,Qiwei Baizhu powder high dose group and middle dose group decreased significantly( P < 0. 05). Compared with the model group,the activity of SOD and GSH-PX in the high dose group and the middle dose group increased significantly,the level of MDA,serum creatinine and urea nitrogen decreased significantly( P < 0. 05);the activity of SOD in the low dose group increased significantly( P < 0. 05),the level of MDA and urine nitrogen decreased significantly( P < 0. 05). Conclusion: Qiwei Baizhu powder can reduce the level of fasting blood glucose,inhibit oxidative stress and improve renal function in the model rats of deficiency of both qi and yin.
