摘 要:通过记录和测量非洲菊单倍体植株的开始生根时间、生根率、根数、根长、苗高和鲜重,比较非洲菊单倍体和二倍体植株在不同培养基中培养的生根效果,研究了单倍体植株在适宜培养基中培养的生根情况。结果表明:培养基和基因型对非洲菊单倍体生根效果存在显著影响,单倍体最佳生根培养基为1/2MS+IBA 0.4 mg/L+NAA 0.3 mg/L。采用1/2MS培养基或添加适宜浓度的植物生长调节剂NAA,可显著提高单倍体的生根率,使单倍体的生根率达77.5%以上。非洲菊单倍体的生根率、根长、株高和鲜重增加速度比二倍体的相应指标都低,单倍体的根系萌发所需时间显著比二倍体的根系萌发时间长。5种单倍体在适宜培养基中培养,根系开始萌发时间12~15 d、根长1.1~1.7 cm、株高2.6~3.7 cm、生根率85%~98.8%、根数8.1~10.1条、鲜重增加速度11.0~12.6 mg/d。By measuring and recording the rooting time, rooting rate, root number, root length, seedling height and fresh weight of gerbera haploid plants, the rooting effects of gerbera haploid and diploid plants in different culture media were compared. The rooting conditions of haploid plants cultured in a suitable medium were tested and screened. The results showed that the medium and genotype had significant effects on the rooting effect of Gerbera hybrida haploid. The best rooting medium of haploid was 1/2 MS+ IBA 0.4 mg/L+NAA 0.3 mg/L. Using 1/2 MS medium or adding appropriate concentration of plant growth regulator NAA, could significantly improve the rooting rate of haploids. The rooting rate of haploids was more than 77.5%. The rooting rate, root length, plant height and fresh weight of gerbera haploids were lower than those of diploids, respectively. The time required for root germination of haploids was also significantly longer than that of diploids. Five species of haploids were cultured in a suitable medium. The roots began to germinate for 12~15 days. The root length was 1.1~1.7 cm. The plant height was 2.6~3.7 cm. The rooting rate was 85%~98.8%. The number of roots was 8.1~10.1, and the rate of increasing fresh weight was 11.0~12.6 mg/d.
