摘 要:目的利用高效液相色谱法对银蒲解毒片中所含绿原酸进行含量测定,并且对实验条件进行优化,确定最佳实验方案。方法采用高效液相色谱法,InertSustainC18色谱柱,流动相为乙腈(A)-0.4%磷酸溶液(B),进样量为10μl,流速为1.0 ml/min,波长为327 nm,柱温为35℃,测定绿原酸的含量。结果绿原酸在选定的色谱条件下,线性回归方程为Y=223.921X-11.142,在1.4~9.8μg/ml范围内线性关系良好(r=0.999 8),RSD=1.52%(n=6)。结论绿原酸为山银花的特征成分,本次实验方法操作简便,准确可靠,可用于控制银蒲解毒片的质量。Objective To determine the content of chlorogenic acid in Yinpu jiedu tablets by high performance liquid chromatography(HPLC), and to optimize the experimental conditions to determine the optimal experimental scheme.Methods The content of chlorogenic acid was determined by HPLC, InertSustainC18 column, acetonitrile-(A)-0.4% phosphoric acid solution(B), 10 μl, flow rate 1.0 ml/min, wavelength 327 nm, column temperature 35℃, the mobile phase consisted of acetonitrile-(A)-0.4% phosphoric acid solution(B), injection volume 10 μl, flow rate 1.0 ml/min, wavelength 327 nm and column temperature 35℃.Results Under the selected chromatographic conditions, the linear regression equation is Y = 223.921 X-11.142.The linear relationship between 1.4 ~ 9.8 g/ml was good(r = 0.999 8),RSD was 1.52%(n = 6). Conclusion Chlorogenic acid is the characteristic component of flos lonicerae. This method is simple, accurate and reliable, and can be used to control the quality of yinpu jiedu tablets.
