摘 要:目的建立小鼠流感性病毒性肺炎模型,检查呼吸道流感病毒感染过程中是否存在肺肠道功能损伤,并探讨加减银翘散的干预作用。方法 BALB/c小鼠36只随机分为正常组、病毒组、加减银翘散组。除正常组外,其余两组滴鼻感染10 LD501/47/FM/H1N1流感病毒液建立小鼠流感病毒性肺炎模型,感染后第5 d取材,HE染色检测肺脏、大肠、小肠、组织病理;ELISA检测肺肠组织流感病毒HA抗原、s IgA、细胞因子、Toll受体及整合素分子水平。结果流感病毒感染后,病毒组小鼠肺组织出现明显的炎性病变外,小肠(十二指肠、回肠、空肠)及大肠(结肠及直肠)均表现出炎症性病变,肺肠组织流感病毒抗原HA水平、细胞因子(TNF-α、IL-6、IL-17),Toll 3、4、9及整合素分子αvβ3、α4水平明显上升,s IgA水平下降,加减银翘散可以减缓肺肠炎症性病变,改善肠粘膜免疫功能。结论流感病毒感染小鼠存在大小肠炎症性病变,且流感病毒能直接在大肠、小肠中复制,加减银翘散能抑制流感病毒感染小鼠肺肠炎性病变,改善肺肠粘膜免疫功能。Objective To establish mouse models of pneumonia induced by influenza virus,and observe occurrence of lung and intestinal damage from influenza virus infection as well as the intervention effect of Modified Yinqiao Powder. Methods Thirty-six BALB/c mice were randomly divided into normal group,virus infected group and Modified Yinqiao Powder group. Mouse models were developed in the other two groups by intranasal infection with 10 LD50 1/47/FM/H1 N1 influenza virus except the normal group. Tissues of the lung,large and small intestines were sampled on the day 5 after infection,and subjected to HE staining to detect the pathologic changes,and ELISA was performed to measure the levels of HA antigen,s IgA,cytokines,Toll receptor and integrin molecules in lung and intestine tissues. Results The lung,small intestines( duodenum,ileum,jejunum) and large intestines( colon and rectum) demonstrated obvious inflammatory lesions in mice following virus infection,with significantly increased levels of HA antigen,cytokines( TNF-α,IL-6,IL-17),Toll 3,4 and 9,integrin αvβ3 and α4,yet decreased s IgA level. Modified Yinqiao Powder resulted in alleviated pulmonary and intestinal inflammatory lesions as well as improved intestinal mucosal immune function. Conclusion Inflammatory lesions occur in the large and small intestines of mice infected by influenza virus,which may replicate directly in the large and small bowels. However,Modified Yinqiao Powder can inhibit the inflammatory lesions of the lungs and intestines induced by influenza virus and improve the mucosal immune function of both lungs and intestines.
