摘  要:以油菜品种“浙大622”为背景材料,将其外植体(子叶柄和下胚轴)分别在含0、10、20、30、40、50和100 mg·L^-1卡那霉素(Kan)或壮观霉素(Spe)的分化培养基中培养,根据其再生芽的生长情况,筛选出最适抗生素浓度.结果表明:外植体的愈伤鲜重、再生芽率和单个外植体再生芽数均随着Kan或Spe浓度的升高而下降;外植体的再生芽白化率随着Kan或Spe浓度的升高而上升,且子叶柄在浓度高于20 mg·L^-1 Kan或30 mg·L^-1 Spe中的再生芽均为白化芽,下胚轴在浓度高于30 mg·L^-1 Kan或20 mg·L^-1 Spe中的再生芽均为白化芽.The seedling cotyledon petioles and hypocotyls of commercial rapeseed(Brassica napus L.)‘Zheda 622'were cultured in differentiation media supplemented with 0,10,20,30,40,50 and 100 mg·L^-1 kanamycin(Kan)or spectinomycin(Spe),respectively.The optimal concentration of antibiotics was screened based on the growth of regenerated shoots.The results showed that fresh weight of callus,rate of regenerated shoots and number of regenerated shoots from single explant decreased significantly with the increase of Kan or Spe concentration while the albino rate of regenerated shoots rose significantly with the increase of Kan or Spe concentration.The regenerated shoots from cotyledon petioles turned albino at concentrations of more than 20 mg·L^-1 Kan or 30 mg·L^-1 Spe,and the regenerated shoots from hypocotyls became albino at concentrations of over 30 mg·L^-1 Kan or 20 mg·L^-1 Spe.
