摘  要:目的观察连梅颗粒对肾阴虚型糖尿病大鼠的降糖作用。方法采用一次性腹腔注射链脲佐菌素(45 mg·kg^-1)+皮下注射甲状腺素(0.2 mg·kg^-1,连续7 d)复制肾阴虚型糖尿病动物模型。将SD雄性大鼠随机分为正常对照组,模型组,连梅颗粒低、中、高剂量组(2.63、5.26、10.52 g·kg^-1),参芪降糖颗粒组(0.625 g·kg^-1),格列齐特片组(16.67 mg·kg^-1),每组8只。灌胃给药,每天1次,连续4周。测定大鼠摄食量、饮水量、体质量;用血糖仪检测大鼠空腹血糖;采用酶联免疫试剂盒测定血清环磷酸腺苷(cAMP)及环磷酸鸟苷(cGMP)水平;灌胃葡萄糖溶液(2 g·kg^-1)进行糖耐量检测,计算血糖曲线下面积(AUC);采用生化试剂盒检测肝糖原与肌糖原含量。结果与正常对照组比较,模型组大鼠空腹血糖值、cAMP水平及cAMP/cGMP比值、摄食量、饮水量、AUC值均显著升高(P<0.05,P<0.01,P<0.001),cGMP水平、体质量、肝糖原及肌糖原含量均显著降低(P<0.01)。与模型组比较,各给药组大鼠的空腹血糖值、cAMP水平及cAMP/cGMP比值、AUC值均显著降低(P<0.05,P<0.01),cGMP水平、肝糖原含量明显升高(P<0.05,P<0.01);连梅颗粒中、高剂量组大鼠的体质量明显升高(P<0.05),而摄食量、饮水量显著降低(P<0.05,P<0.01);连梅颗粒高剂量组的肌糖原含量显著升高(P<0.05)。结论连梅颗粒可降低肾阴虚型糖尿病大鼠血糖,可能与调节机体环核苷酸系统、促进肝糖原及肌糖原合成有关。Objective To observe the hypoglycemic effects of Lian Mei Keli on diabetic rat models with kidney-yin deficiency.Methods The diabetic model rats of kidney-yin deficiency were induced by intraperitoneal injection of streptozotocin(45 mg·kg^-1)in combination with subcutaneous injection of thyroxine(0.2 mg·kg^-1,7 d).Male SD rats were randomly divided into Healthy control group,Model group,low,medium,high dosages of Lian Mei Keli groups(LMKL 2.63,5.26,10.52 g·kg^-1),Shen Qi Jiangtang Keli group(SQJTKL 0.625 g·kg^-1),and Gliclazide tablets group(Gliclazide 16.67 mg·kg^-1).There are 8 rats in each group with intragastric administration once daily for 4 weeks.The food consumption,water consumption and body weight of rats were determined.Fasting plasma glucose was determined by glucometer,the levels of cAMP and cGMP in serum were determined by ELISA.The glucose tolerance after glucose(2 g·kg^-1)by intragastric administration was determined and the AUC was calculated.Levels of hepatic glycogen and muscle glycogen were determined by Glycogen Assay Kit.Results Compared with the Healthy control group,blood glucose,the levels of cAMP,the ratio of cAMP/cGMP,food consumption,water consumption and AUC significantly increased(P<0.05,P<0.01,P<0.001).The level of cGMP,body weight,hepatic glycogen and muscle glycogen significantly decreased(P<0.01).Compared with the Model group,blood glucose,the levels of cAMP,the ratio of cAMP/cGMP,AUC significantly decreased(P<0.05,P<0.01).The levels of cGMP,hepatic glycogen and muscle glycogen significantly increased(P<0.05,P<0.01).Body weight of rats in Lian Mei Keli medium and high dosage groups significantly increased(P<0.05),but the food consumption and water consumption significantly decreased(P<0.05,P<0.01);the level of muscle glycogen of rats in Lian Mei Keli high dosage group significantly increased(P<0.05).Conclusion Lian Mei Keli could lower blood glucose of diabetic model rats with kidney-yin deficiency.Its mechanism may be related to regulating the cyclic nucleotide system and
