木豆叶提取物对H2O2诱导损伤HUVEC的影响

摘  要:目的研究木豆叶提取物对H2O2诱导损伤的人脐静脉内皮细胞株(HUVEC)的影响,为其抗动脉粥样硬化(Atherosclerosis,AS)提供技术支撑。方法制备各组含药血清。体外培养HUVEC,设立空白组、模型组、VitC组(0.08 g·kg^-1,15%含药血清)、木豆叶低剂量组(2.5 g·kg^-1,5%含药血清)、木豆叶中剂量组(2.5 g·kg^-1,10%含药血清)、木豆叶高剂量组(2.5 g·kg^-1,15%含药血清)。采用H2O2损伤HUVEC,给予相应药物共培养24 h。MTT法检测细胞活力,AnnexinV-FITC/PI双染法检测细胞凋亡,微量酶标法检测乳酸脱氢酶(Lactic Dehydrogenase,LDH)含量,硝酸还原酶法检测一氧化氮(Nitric Oxide,NO)含量,酶联免疫吸附法(Elisa)检测内皮素-1(Endothelin-1,ET-1)含量。结果与模型组比较,VitC组、木豆叶中剂量组细胞活力明显提高(P<0.01);VitC组、木豆叶中、高剂量组的细胞凋亡减少,木豆叶中浓度组逆转细胞凋亡效果最好;VitC组、木豆叶低剂量组的LDH含量显著提高,木豆叶高剂量组LDH含量显著降低(P<0.05或P<0.01);木豆叶低、高剂量组NO含量显著提高(P<0.01);木豆叶低、中、高剂量组ET-1含量显著降低(P<0.01)。结论木豆叶提取物含药血清具有抗H2O2诱导的AS内皮损伤作用。Objective To study the effect of extracts of Cajanus cajan leaf(CCL)on human umbilical vein endothelial cells(HUVECs)in vitro.Methods The serum containing correspondent drugs were prepared.HUVECs were cultured in vitro.Cells were divided into blank control group,model group,Vitamin C group(0.08 g·kg^-1,15%drugcontaining serum),CCL low dosage group(2.5 g·kg^-1,5%drug-containing serum),CCL medium dosage group(2.5 g·kg^-1,10%drug-containing serum),CCL high dosage group(2.5 g·kg^-1,15%drug-containing serum).Hydrogen peroxide was used to induce damage in HUVECs and corresponding drugs were given for 24 hours.The MTT method was used to detect the cell viability.AnnexinV-FITC/PI double staining method was used to detect the changes of apoptosis.The micro-enzyme-linked method was used to detect the release of lactate dehydrogenase(LDH).The nitrate reductase activity method was used to detect the content of Nitric oxide.The enzyme-linked immuno sorbent assay was used to detect the content of endothelins-1(ET-1).Results The results reveal that the viability of HUVECs was improved when treated with CCL medium dosage and VitC(P<0.01).The apoptosis of HUVECs was decreased when treated with VitC,CCL medium and high dosages.CCL medium dosage showed the best effect.The release of LDH was significantly improved when treated with VitC and CCL low dosage,the release of LDH was significantly decreased when treated with CCL high dosage(P<0.05 or P<0.01).The contents of Nitric oxide were significantly improved when treated with CCL low and high dosages(P<0.01).The contents of ET-1 were significantly decreased when treated with CCL low,medium and high dosages(P<0.01).Conclusion It showed that the CCL extract containing serum has inhibitory effects on damage of HUVECs in atherosclerosis induced by hydrogen peroxide.
