紫草婴儿软膏对湿疹的治疗作用及机制

摘　　要：目的研究紫草婴儿软膏对动物实验性湿疹模型的治疗作用并探讨其作用机制,为其临床应用提供实验依据。方法复制豚鼠卵清蛋白(ovalbumin,OVA)激发变态反应性湿疹模型和小鼠2,4-二硝基氯苯(2,4-dinitrochlorobenzene,DNCB)诱导的婴幼儿湿疹样模型,分别评价紫草婴儿软膏对实验性湿疹的治疗作用;用TUNEL法检测紫草婴儿软膏对皮损组织角质形成细胞(keratinocyte,KC)细胞凋亡的影响。结果0.5,1和2 g/kg的紫草婴儿软膏均能降低湿疹模型动物的皮损红斑、水肿、抓痕及总评分。病理学检测表明,1,2 g/kg的紫草婴儿软膏能改善湿疹模型动物皮肤病变程度(P<0.05);TUNEL检测结果显示,紫草婴儿软膏能明显增加阳性细胞数(P<0.05)。结论紫草婴儿软膏对小鼠实验性湿疹有明显治疗作用,其机制与促进角质形成细胞凋亡密切相关。Objective To study the therapeutic effects of Lithospermum Ointment/Zi Cao baby ointment on Eczema,and to provide experimental basis for its clinical application.Me thods The model of skin allergic eczema induced byovalbumin(OVA)in guinea pigs and the infantile eczema like model in mice induced by 2,4-dinitrochlorobenzene(DNCB)were duplicated to evaluate the therapeutic effects of Zi Cao baby ointment on experimental eczema,respectively.TUNEL method was used to investigate the effects of Zi Cao baby ointment on the apoptosis of keratinocytes and lymphocyte.Re s ults Zi Cao baby ointment at 0.5,1 and 2 g/kg could reduce the degrees of erythema,edema,scratch and total score of eczema model animals;Pathological examination showed that Zi Cao baby ointment of 1,2 g/kg ccould improve the degree of skin lesions in the guinea pigs;Zi Cao baby ointment at 1,2 g/kg could reduce the auricular swelling of the mice,and reduce the score of skin lesions,improve the degree of skin lesions in the mice.Zi Cao baby ointmen increased the numbers of positive cells in the TUNEL test.Conclus ion The results showed that Zi Cao baby ointment had obvious therapeutic effects on experimental eczema in the animal models and the possible mechanisms were closely related to the promotion of the apoptosis of keratinocytes(KC).
