摘 要:目的探讨TGF-1β和MMP-2/MMP-9mRNA在PQ中毒鼠肺中的表达及Ac-SDKP保护作用。方法实验分为Ac-SDKP治疗组(60只),对照组(C30只)、百草枯中毒组(PQ60只)。PQ组按照25 mg/kg标准给予百草枯灌胃造模,C组灌胃生理盐水。Ac-SDKP治疗组:造模后1 h腹腔埋入含Ac-SDKP[800μg/(kg·d)]的微量缓释泵;PQ组、C组分别予生理盐水。以6 h、12 h、1 d、3 d、5 d、7 d为研究点处死实验鼠,C组每次5只,其它组每次10只。对实验鼠肺泡炎症程度、间质水肿、肺不张和肺纤维化形成分别进行评分,实验鼠肺组织TGF(转化生长因子)-1β和MMP-2/MMP-9mRNA的表达检测分别采用酶联免疫法(ELISA)方法和逆转录-聚合酶链反应(RT-PCR)方法检测。结果与对照组相比,PQ组肺组织损伤明显,TGF-1β的表达升高,MMP-2/MMP-9mRNA的表达上调(P <0.05),Ac-SDKP组能抑制这种变化;TGF-1β与MMP-2/MMP-9 mRNA表达呈正相关(r1=0.71,r2=0.87,P <0.01)。结论 Ac-SDKP可能也是通过对TGF-1β和MMP-2/MMP-9 mRNA基因表达的调节,来保护百草枯中毒所致肺损伤。Objective To investigate the expressions of TGF-1β and MMP-2/MMP-9 in the lungs of PQ poisoned rats and the protective effect of Ac-SDKP. Me thods One hundred and fifty SD rats were randomly divided into 3 groups: normal control group(C) with 30 rats, paraquat poisoning group(PQ) with 60 rats, and paraquat poisoning group with 60 rats. The PQ group rats were given paraquat by gastric perfusion(25 mg/kg);The C group rats were given saline by gastric perfusion. Ac-SDKP treatment group was made by implanting a micropump containing Ac-SDKP[800 ug/(kg·d)] into the abdominal cavities one hour after modeling. The PQ and C group rats were given saline. Mice were sacrificed at 6 h,12 h,1 d,3 d,5 d and 7 d,5 for each time in C group,and 10 in other groups. Histological changes of lung tissues under light microscope were observed.The TGF-1βprotein was evaluated by western blot and the expressions of MMP-2 and MMP-9 mRNA in the lung tissues of rats were measured by RT-PCR. Re s ults Ac-SDKP could reduce the expressions of TGF-1β、MMP-2 and MMP-9, which were induced by PQ. The expression of TGF-1βwas positively correlated with MMP-2’s(r1=0.71, P < 0.01), and positively correlated with MMP-9’s(r2=-0.87,P < 0.01). Conclus ion Ac-SDKP may protect against the lung injuries caused by paraquat poisoning by regulating the gene expressions of TGF-1 beta and MMP-2/MMP-9.
