摘  要:[目的]本试验旨在探究早期能量限饲与后期能量补偿对湖羊生长性能、肝脏组织中Hippo信号通路核心基因和AMPK蛋白表达的影响。[方法]选择32只系谱背景清楚、体质量相近的3月龄雄性湖羊羔羊,随机均分为对照组和补偿生长组。试验分为能量限饲(60 d)和补偿(90 d)2个阶段,能量限饲阶段对照组和试验组饲喂日粮的代谢能分别为11.64和6.84 MJ·kg^-1,在能量补偿阶段日粮的代谢能均为12.86 MJ·kg^-1。饲养结束后屠宰,测定,再通过RT-qPCR和Western blot检测肝脏组织中Hippo通路核心基因和AMPK蛋白的表达变化。[结果]在能量限饲阶段,与对照组相比,能量限饲显著降低了补偿组湖羊的肝脏质量和体质量(P<0.05),湖羊肝脏组织中Hippo通路核心基因和YAP1蛋白的表达量显著降低(P<0.05),而AMPK的mRNA和蛋白磷酸化水平显著升高(P<0.05)。在能量补偿阶段,补偿组湖羊的肝脏质量和体质量、湖羊肝脏组织中Hippo通路核心基因(除YAP1外)的表达量和AMPK蛋白的磷酸化水平均与对照组差异不显著。[结论]湖羊在适当水平的能量限饲后进行补偿处理具有补偿生长现象,Hippo通路和AMPK蛋白可能共同参与不同营养水平下对湖羊肝脏发育的调节。[Objectives]This study was conducted to elucidate the growth performance,the expression levels of Hippo signaling pathway components and AMP-activated protein kinase(AMPK)in the liver of Hu sheep under caloric restriction and compensation situations.[Methods]Thirty-two ram lambs were randomly assigned into control group and compensation growth group(three months of age,n=16)based on the body weight.Caloric restriction period(60 d)and caloric compensation period(90 d)were designed.At caloric restriction period,the animals in the control and the caloric restriction group were fed with normal diet(11.64 MJ·kg^-1)and calorie restriction diet(6.84 MJ·kg^-1),respectively.Subsequently,at caloric compensation period,animals in both groups were fed with regular diet(12.86 MJ·kg^-1)for additional three months.Lambs were slaughtered at the end of caloric restriction and compensation period,the body weights were measured and the liver tissues were collected which were used to analyze the expressions of Hippo signaling pathway components and AMPK by real-time PCR and Western blot.[Results]During the caloric restriction period,the body and liver weights of the lamps significantly decreased in the caloric restriction group(P<0.05).The expression levels of Hippo pathway components,and the translation levels of YAP1 were significantly reduced in the caloric restriction group than those in the control group(P<0.05),while the mRNA level of AMPK and protein level of pAMPK significantly increased under caloric restriction(P<0.05).After the caloric re-alimentation,no significant differences in growth or tested hepatic elements were detected.[Conclusions]The Hippo pathway and AMPK in liver tissues might effectively participate in the metabolic regulation under temporal energy restriction and subsequent re-alimentation.
