摘  要:利用HPLC法测定3种不同花色木本曼陀罗不同部位中莨菪碱和东莨菪碱的含量,比较不同花色不同部位中2个成分的含量分布。采用InertSustain C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-0.07 mol/L磷酸钠溶液(含0.017 5 mol/L十二烷基硫酸钠)(50∶100)为流动相,梯度洗脱,流速1.0 mL/min,检测波长216 nm,柱温35℃。结果表明,不同花色木本曼陀罗不同组织部位中莨菪碱和东莨菪碱含量差异较大。红花木本曼陀罗中莨菪碱含量最高部位是主根,为0.066%,东莨菪碱含量最高部位是花苞,为0.114%;黄花木本曼陀罗中莨菪碱和东莨菪碱含量最高部位均为嫩叶,分别为0.156%和0.287%;白花木本曼陀罗中莨菪碱含量最高部位是花苞,为0.058%,东莨菪碱含量最高部位是一年生木质化茎,为0.433%。该研究明确了3种花色木本曼陀罗不同部位中莨菪碱和东莨菪碱含量差异,为上述资源植物的开发利用提供了实验依据。To determinate the contents of hyoscyamine and scopolamine in different parts of three colorful Brugmansia Arborea by HPLC.And to compare the content distribution of the above two flavones in different parts of three colorful plants.HPLC analysis was performed on a InertSustain C18 column(4.6 mm×250 mm,5 μm).The mobile phase was acetonitrile-sodium phosphate(0.07 mol/L) containing 0.017 5 mol/L lauryl sodium sulfate(50∶100) with gradient elution.The flow rate was 1.0 mL/min,the detection wavelength was 216 nm,and the column temperature was 35 ℃.The result was as follow:The contents of hyoscyamine and scopolamine in different parts of different colorful Brugmansia Arborea varied greatly.The highest content of hyoscyamine(0.066%) was found in main roots,and the highest content of scopolamine(0.114%) was found in flower buds of Brugmansia Arborea with red flower.The highest content of hyoscyamine(0.156%) and scopolamine(0.287%) were found in young leaves of Brugmansia Arborea with yellow flower.The highest content of hyoscyamine(0.058%) was found in flower buds,and the highest content of scopolamine(0.433%) was found in annual woody stems of Brugmansia Arborea with white flower.The content distributions of hyoscyamine and scopolamine in different parts of three colorful Brugmansia Arborea was determinated.The result of the study provides experimental reference for the utilization of Brugmansia Arborea as scopolamine resource plant.
