摘 要:目的:探讨核因子κB受体活化因子配体(RANKL)和肿瘤坏死因子-α(TNF-α)在牙周炎大鼠正畸性牙根吸收中牙周膜压力侧的表达。方法:40只SD雄性大鼠随机分为牙周炎组和对照组,每组20只。近移上颌第一磨牙,在加力后0、3、7、14 d各处死5只,苏木精-伊红(HE)染色观察牙根吸收、抗酒石酸酸性磷酸酶(TRAP)染色测破牙骨质细胞数目、免疫组织化学法检测压力侧牙周膜中RANKL、TNF-α的表达。结果:牙根吸收随加力时间延长而增加,且加力7、14 d时牙周炎组高于对照组(P<0.05);加力3、7、14 d时牙周炎组RANKL、TNF-α表达高于对照组(P<0.05)。结论:牙周炎可加重牙根吸收,可能与牙周炎症状态下牙周膜中RANKL、TNF-α表达上调有关。Objective: To investigate the expression of receptor activator of nuclear factor-κB ligand(RANKL) and tumor necrosis factor-α(TNF-α) in periodontal ligament of compressive side during orthodontic root resorption of rats with periodontitis. Methods: 40 male SD rats were randomly divided into periodontitis group and control group(n=20). The maxillary first molar was pulled mesially in the 2 groups. 5 rats were executed at each time after activation of pull-spring for 0, 3, 7 and 14 d respectively in the 2 groups. HE staining was used to observe the root resorption, and the number of odontoclasts was measured by tartrate acid phosphatase(TRAP) staining. The expression of RANKL and TNF-α in the periodontal ligament of compressive side was detected by immunohistochemistry(IHC). Results: The root resorption increased after mechanical loading, and was significantly more severe in the periodontitis group than that in the control group at 7 th and 14 th d(P<0.05). The RANKL and TNF-α expression in the periodontitis group was significantly higher than that in the control group at 3 rd,7 th and 14 th d(P<0.05). Conclusion: Periodontitis aggravates the orthodontic side effect of root resorption, which may be related to the upregulation of RANKL and TNF-α expression in the periodontal ligament during periodontal inflammation.
