摘 要:目的探讨去除尼古丁和焦油的香烟烟雾提取物(fCSE)与尼古丁对血清睾酮和勃起功能影响的差异及相关机制.方法选取30只SD雄性大鼠,随机分为三组:PBS组、fCSE组及尼古丁组;8周后利用放射免疫法检测血清睾酮(T)水平,电刺激阴茎海绵体神经测定平均动脉压(MAP)、阴茎海绵体内压峰值(MICP)、海绵体充盈率(CFR)及达到阴茎海绵体内压峰值的刺激时间Tmax等指标评估各组大鼠的勃起功能.结果fCSE组、尼古丁组血清睾酮水平较PBS组下降[(3.22±1.11)ng/mL、(2.58±0.81)ng/mL、(5.37±1.43)ng/mL];fCSE组、尼古丁组勃起功能学指标MAP及Tmax较PBS组升高,MICP[(58.99±10.76)mmHg,(54.79±8.54)mmHg]及CFR[(0.89±0.44)、(0.77±0.82)]较PBS组[MICP(85.92±6.36)mmHg,CFR(2.14±0.44)]下降,差异有统计学意义(P<0.05).结论去除尼古丁和焦油的香烟烟雾提取物和尼古丁对大鼠血清睾酮水平和阴茎勃起功能的下降与氧化应激损伤相关,但尼古丁在勃起功能和激素水平下降更明显.Objective To evaluate the differences and mechanisms of nicotine-and tar-free cigarette smoke extract(fCSE)on plasma testosterone level and erectile function in rats.Methods Thirty SD male rats were randomly divided into three groups:the control group(PBS group),the fCSE group and the nicotine group.After 8 weeks,the levels of plasma testosterone(T)were measured with radioimmunoassay by electrostimulating of the the cavernous nerve,MAP(mean arterial pressure),MICP(maximal intracavemous pressure),CFR(cavernosal filling rate)and Tmax(the time of stimulating MICP happened),were measured with radioimmunoassay to evaluate the erectile function of rats.Results Compared with the control group[T(5.37±1.43)ng/mL],the plasma testosterone of fCSE group and the nicotine group[T(3.22±1.11)ng/mL T(2.58±0.81)ng/mL]decreased obviously;The MAP[(105.15±4.43)mmHg,(108.99±6.76)mmHg]and Tmax[(42.90±8.56)s,(43.56±2.43)s]in fCSE group and nicotine group were higher than those in PBS group[MAP(100.99±6.95)mmHg,Tmax(29.90±5.78)s];MICP[(58.99±10.76)mmHg,(54.79±8.54)mmHg]and CFR[(0.89±0.44),(0.77±0.82)]were significantly lower than those of PBS group[MICP(85.92±6.36)mmHg,CFR(2.14±0.44)].Compared with the control group,the difference was significanl(P<0.05),while the difference between nicotine group and PBS was more obvious(P<0.05).Conclusions The decreases of nicotine-and tar-free cigarette smoke extract and nicotine in plasma testosterone level and penile erectile function in rats are related to oxidative stress injury,but the decrease of nicotine is more obvious.
