养血润燥法对银屑病皮损中IRF-4表达的影响

摘　　要：目的:探讨养血润燥法治疗银屑病的作用机制,研究以养血润燥法为组方原则的润肤止痒乳对银屑病患者皮损中干扰素调节因子-4(IRF-4)的影响。方法:72只小鼠造模并分为正常对照组、空白模型组、润肤止痒乳高浓度组(0.48 g/mL)、中浓度组(0.24 g/mL)、低浓度组(0.12 g/mL)以及他扎罗汀组。在第3天及第8天取小鼠背部脱毛区皮肤,以免疫组化检测IRF-4的表达。结果:外观观察与HE染色证实造模成功。比较各组小鼠的累积光密度(IOD),结果显示:所有组与空白模型组比较,差异均有统计学意义(P<0.05);润肤止痒乳高浓度组、润肤止痒乳低浓度组及他扎罗汀组IRF-4的表达高于正常对照组(P<0.05),而与润肤止痒乳中浓度组比较差异无统计学意义(P>0.05);润肤止痒乳中浓度组中IRF-4表达低于润肤止痒乳高浓度组及低浓度组(P<0.05);润肤止痒乳高、中、低浓度组与他扎罗汀组比较差异无统计学意义(P>0.05)。结论:润肤止痒乳治疗银屑病临床疗效确切,并且抑制了银屑病皮损中IRF-4的表达,从而证实了玄府-肺理论在银屑病治疗中的指导作用,也探讨了养血润燥法治疗银屑病的作用机制。Objective:To study the Yangxue Runzao Method on psoriasis.To discuss the efficacy of the Runfu Zhiyang Ru which based on Yangxue Runzao Method and Xuanfu-lung Theory,and the expression of interferon reguLatory factor-4(IRF-4)in patients of psoriasis.Methods:72 mice were divided into six groups:normal group,control group,the Runfu Zhiyang Ru low(0.12 g/mL),middle(0.24 g/mL),high(0.48 g/mL)dose group and the tazarotene group.Taking photos of mice,everyday.And cut off the central lesion of the skin on the 3 rd day and 8 th day.Make sure the molding is successfuL by appearance assessment and HE staining method.Ivestigate the expression of interferon reguLatory factor-4(IRF-4)through immunohistochemical method.Results:Analyze the IOD between each groups.In the control group,the expression of IRF-4 is higher than other groups(P<0.05).The expression of IRF-4 in the normal group is lower than it in the Runfu Zhiyang Ru low,highdose group and the tazarotene group(P<0.05),and which was not statistically significant compared with the Runfu Zhiyang Ru middle dose group(P>0.05).The expression of IRF-4 in the Runfu Zhiyang Ru middle dose group is statistically significant compared with it in the Runfu Zhiyang Ru low and highdose group.In the tazarotene group,the expression of IRF-4,which was not statistically significant compared with the Runfu Zhiyang Ru low,middle and high dose group(P>0.05).The study show that Runfu Zhiyang Ru and tazarotene both can supress the expression of IRF-4.The Runfu Zhiyang Ru of middle dose group was equivalent to or better than the tazarotene group and is the best concentration of all.Conclusion:Runfu Zhiyang Ru has great curative effect on psoriasis,and decrease the expression of IRF-4.Moreover,this study not only proved the concrete mechanism of Yangxue Runzao Method in treatment of psoriasis,but also demonstrate the guiding role of Xuanfu-lung Theory in the therapy of psoriasis.
