摘 要:目的:初步探讨葛根散对酒伤勃起功能障碍(erectile dysfunction,ED)大鼠阴茎平滑肌组织β-链蛋白(β-catenin)mRNA及蛋白表达的影响。方法:将SD大鼠随机分为正常组,模型组,葛根散低、中、高剂量组(0.05 g·10 g^-1、0.1 g·10 g^-1、0.2 g·10 g^-1)5组。用荧光定量聚合酶链式反应法(Real-time PCR)检测葛根散对酒伤ED大鼠阴茎平滑肌组织β-catenin mRNA表达的影响情况;用蛋白免疫印迹法(Western blot)检测葛根散对酒伤ED大鼠阴茎平滑肌组织β-catenin蛋白表达的影响情况。结果:与正常组比较,模型组β-catenin mRNA及蛋白表达水平显著下降(P<0.05)。与模型组比较,葛根散中、高剂量组β-catenin mRNA表达水平显著上升,葛根散低、中、高剂量组β-catenin蛋白表达水平显著上升(P<0.05)。且葛根散高剂量组β-catenin蛋白相对表达水平与正常组比较无统计学差异(P<0.05)。结论:葛根散对酒伤ED大鼠阴茎平滑肌组织具有保护作用,其作用机制可能与调节酒伤ED大鼠阴茎平滑肌组织β-catenin mRNA和蛋白表达水平有关。Objective:To investigate the effect of gegen powder on the expression ofβ-catenin mRNA and protein in penile smooth muscle of rats with erectile dysfunction(ED).Methods:SD rats were randomLy divided into 5 groups:normal group,model group,gegen powder low,medium and high dose group(0.05 g·10 g^-1,0.1 g·10 g^-1,0.2 g·10 g^-1).The expression ofβ-catenin mRNA in penile smooth muscle of ED rats was detected by real-time PCR.The expression ofβ-catenin mRNA in penile smooth muscle of ED rats was detected by Western blot.Expression ofβ-catenin protein in penile smooth muscle tissue of ED rats.Results:Compared with the normal group,the expression ofβ-catenin mRNA and protein in the model group was significantly decreased(P<0.05).Compared with the model group,the expression ofβ-catenin mRNA was significantly increased in the middle and high doses of gegen powder,and the expression ofβ-catenin protein in the low,middle and high doses of Pueraria was significantly increased(P<0.05).The relative expression ofβ-catenin protein in the high dose group of gegen powder was not significantly different from that in the normal group(P<0.05).Conclusion:Gegen powder has protective effect on penile smooth muscle tissue of ED rats,and its mechanism may be related to the regulation ofβ-catenin mRNA and protein expression in penile smooth muscle tissue of rats with ED.
