摘 要:目的:探讨丹红注射液对大鼠急性脑梗死后血管新生的影响。方法:随机选取20只SD大鼠为空白组,20只SD大鼠为假手术组,60只SD大鼠采用线栓法制作永久性脑梗死模型,造模成功后随机分为模型组、丹红治疗组、丁苯酞治疗组,每组20只,丹红治疗组予丹红注射液腹腔注射(4.2 mL/kg),丁苯酞治疗组予丁苯酞氯化钠注射液腹腔注射(10 mL/kg),模型组、假手术组、空白组均予等量生理盐水腹腔注射,1次/d,分别连续给药7 d、14 d。于给药第7天、14天给予过量麻醉药处死,每次每组大鼠10只,采用免疫组化法测定血管内皮生长因子(VEGF)表达及微血管密度(MVD)变化,用TTC染色法测定大鼠脑梗死体积。结果:丹红治疗组、丁苯酞治疗组大鼠VEGF表达明显高于空白组、假手术组、模型组(P<0.05);丹红治疗组、丁苯酞治疗组缺血区MVD明显多于模型组(P<0.05);丹红治疗组、丁苯酞治疗组大鼠脑梗死体积明显小于模型组(P<0.05)。结论:丹红注射液能促进大鼠脑梗死后新生血管的形成,缩小梗死体积,其机制可能为上调缺血脑组织VEGF的表达,增加缺血半暗带内血管密度。Objective: To investigate the effect of Danhong injection on angiogenesis after acute cerebral infarction in rats. Methods: 20 SD rats were randomly selected as the blank group, 20 SD rats were used as the sham operation group, and 60 SD rats were used to make the permanent cerebral infarction model by thread embolization. After successful modeling, the rats were randomly divided into the model group and Danhong treatment group, and butylphthalide treatment group, with 20 rats in each group. Danhong treatment group was intraperitoneally injected with Danhong injection(4.2 mL/kg), butylphthalide treatment group was intraperitoneally injected with sodium butylphthalide chloride injection(10 mL/kg), model group, sham operation group and blank group were intraperitoneally injected with the same amount of normal saline once a day for 7 days and 14 days respectively. Excessive anesthetics were sacrificed on the 7 th and 14 th day after administration, and 10 rats in each group were used to determine the expression of vascular endothelial growth factor(VEGF) and microvessel density(MVD) by immunohistochemistry. The volume of cerebral infarction was determined by TTC staining. Results: The expression of VEGF in Danhong treatment group and butylphthalide treatment group was significantly higher than that in blank group, sham operation group and model group(P<0.05), and MVD in ischemic area in Danhong treatment group and butylphthalide treatment group was significantly higher than that in model group(P<0.05). The volume of cerebral infarction in Danhong treatment group and butylphthalide treatment group was significantly lower than that in model group(P<0.05). Conclusion: Danhong injection can promote the formation of neovascularization and reduce infarct volume after cerebral infarction in rats. The mechanism may be to increase the expression of vascular endothelial growth factor in ischemic brain tissue and increase the vascular density in ischemic penumbra.
