摘 要:目的评价基于多色荧光探针熔解曲线技术的结核分枝杆菌新型间隔区寡核苷酸分型技术(McSpoligotyping)的临床应用价值。方法在2013-2014年间,分别从河南省五个片区的10个市或县收集肺结核患者痰标本中分离培养的结核分枝杆菌502株,采用McSpoligotyping和传统的间隔区寡核苷酸分型技术(Spoligotyping)进行分型;比较两者的结果,对两种分型结果不一致的菌株进行基因测序。结果 502株结核分枝杆菌中McSpoligotyping和Spoligotyping检测结果一致的有485株(96.60%),不一致的有17株(3.39%);不一致菌株经基因测序,McSpoligotyping分型有15株(88.24%)与测序结果一致,Spoligotyping分型有2株(11.76%)与测序结果一致。结论 McSpoligotyping作为新型的基因分型方法,较传统Spoligotyping技术更为简便、快速,可以替代传统的Spoligotyping检测技术用于结核病的分子流行病学研究。Objective To investigate a new strain differentiation method of Spoligotyping, base on multiplex fluorescence probe melting curve analysis(McSpoligotyping)for differentiating clinical strains of Mycobacterculosis tuberculosis. Methods From 2013 to 2014,five hundred and two strains of M.tuberculosis collected from pulmonary tuberculosis patients in five districts of Henan province were differentiated using traditional Spoligotyping and McSpoligotyping simultaneously,and the strains were sequenced that had disaccord results by two methods. Results Of502 strains,485(96.60 %)strains had same results by two methods,17(3.39 %)strains were sequenced with the disacoord results.Of 17 strains sequenced,15(88.24 %)strains had same results as Mcspoligotyping and 2(11.76 %)strains had same results as Spoligotyping. Conclusions Mcspoligotyping is rapid and sample as a new method,that should replace the traditional Spoligotyping as molecular epidemiological investigation tool for tuberculosis prevention.
