摘 要:目的观察石斛合剂对糖尿病模型大鼠脂代谢相关基因表达及血脂水平的影响。方法选取SPF级雌性Wistar大鼠50只,采用随机数字表法分为空白组10只、造模组40只,空白组给予基础饲料喂养,造模组给予高脂、高糖饲料喂养,在此基础上造模组腹腔注射链脲佐菌素,将模型复制成功的32只大鼠随机分为模型组、阳性对照组、治疗组,各组分别予生理盐水、二甲双胍混悬液、石斛合剂水煎液灌胃。灌胃24 d后,检测各组大鼠血清甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、总胆固醇(CHOL)水平;取肝脏组织,进行基因芯片检测,并采用免疫组化法检测肝脏组织中β-catenin、CROT的表达量,在光镜下观察肝脏病理改变。结果与空白组比较,模型组大鼠肝脏组织中有1 339个已知功能基因发生异常表达,差异有统计学意义(P<0.05),经过石斛合剂治疗后,有近千个基因表达量恢复了正常,约380个已知功能基因尚未恢复正常;石斛合剂能明显下调WISP2等基因的表达,上调FABP5、MLXIP、SLC10A6等基因的表达,使造模后异常表达的基因如ATP13A1、CROT、ACOT2、LRP5等基因表达恢复正常。与模型组比较,治疗组大鼠血清HDL水平、肝脏组织中CROT表达量均明显升高,血清LDL、TG、CHOL水平及肝脏组织中β-catenin表达量均明显降低,差异有统计学意义(P<0.01)。结论石斛合剂降血脂的作用机制与改善多条基因通路的表达有关,可通过调节WNT、PPARβ/δ等信号通路,起到调节脂代谢、保护肝脏的作用。Objective To observe the effects of Shihu Heji( 石斛合剂) on lipometabolic related regulatory gene expression and blood-lipid levels in diabetes model rats.Methods Fifty SPF female Wistar rats were divided into blank group with 10 rats and establishing model group with 40 rats according to random number table method.The blank group was fed with basic feed,while the establishing model group was fed with high-fat and high-sugar feed.On this basis,the establishing model group was intraperitoneally injected with streptozotocin,and 32 rats successfully replicating the model were randomly divided into the model group,the positive control group and the treatment group,and each group was given normal saline,metformin suspension and Shihu Heji water decoction by gavage. After 24 days of gavage,serum triglyceride( TG),high-density lipoprotein( HDL),low-density lipoprotein( LDL) and total cholesterol( CHOL) levels were detected in each group.The liver tissues were taken for gene chip detection,and the expressions of β-catenin and CROT in the liver tissues were detected by immunohistochemistry,and the pathological changes of the liver were observed under the light microscope. Results Compared with the blank group,1 339 known functional genes were abnormally expressed in the liver tissue of the model group,the difference was statistically significant( P<0.05).After treatment with Shihu Heji,nearly 1 000 gene expression levels returned to normal,about 380 known functional genes have not returned to normal.Shihu Heji could significantly down-regulate the expression of WISP2 and other genes,up-regulate FABP5,MLXIP,SLC10 A6 and so on. The abnormally expressed genes after modeling such as ATP13 A1,CROT,ACOT2 and LRP5 returned to normal. Compared with the model group,the expression of HDL in serum level and CROT in liver tissue increased significantly in the treatment group,while the expression of LDL,TG,CHOL in serum level and β-catenin in liver tissue decreased significantly,and the difference was statistically significant(
