摘 要:目的探讨葛根异黄酮对去势小鼠前列腺增生的影响及其机制。方法小鼠麻醉切除双侧睾丸后,皮下注射丙酸睾酮7 mg·kg^-1·d^-1建立前列腺增生模型。按体重将雄性小鼠随机分为6组:正常组、模型组、对照组(非那雄胺1.4 mg·kg^-1·d^-1)和低、中、高3个剂量实验组(葛根异黄酮50,100,200mg·kg^-1·d^-1)。造模第2天,各组灌胃蒸馏水、非那雄胺和不同剂量的葛根异黄酮,持续给药4周。用酶联免疫吸附法检测血清中5α-还原酶(5AR)的活性和前列腺组织的肿瘤坏死因子(TNF-α)与白细胞介素-8(IL-8)的含量,以分光光度法检测血清中超氧化物歧化酶(SOD)和丙二醛(MDA)的含量。结果正常组、模型组、对照组和低、中、高3个剂量实验组的5AR活性分别为(2.46±0.06),(2.64±0.10),(2.51±0.08),(2.56±0.03),(2.48±0.05)和(2.53±0.04)U·L^-1;上述这6组前列腺组织中TNF-α的含量分别为(102.96±7.17),(118.56±5.16),(100.59±5.46),(104.11±2.68),(103.03±3.52)和(102.54±9.95)pg·m L^-1;上述这6组的IL-8含量分别为(9.07±0.54),(10.81±0.67),(9.46±0.74),(9.78±0.31),(9.28±0.55)和(9.21±0.73)pg·m L^-1;上述这6组的SOD活力分别为(240.76±27.81),(173.03±14.48),(249.21±18.12),(218.55±31.20),(255.53±19.52)和(219.06±16.90)U·m L^-1;上述这6组的MDA含量分别为(7.64±1.16),(11.58±0.68),(9.51±1.06),(9.23±1.12),(8.91±0.82)和(9.90±1.50)nmol·m L^-1。上述指标:模型组与正常组比较,差异均有统计学意义(均P<0.01);3个剂量实验组和对照组与模型组比较,除低剂量实验组的5AR外,其余指标和组间差异均有统计学意义(P<0.05或P<0.01)。结论葛根异黄酮对小鼠前列腺增生具有一定的抑制作用,其机制可能通过降低5AR活性、降低TNF-α和IL-8含量、提高SOD抗氧化酶活力和清除MDA脂质过氧化产物等作用有关。Objective To investigate the effect of Pueraria isoflavones on prostatic hyperplasia in ovariectomized mice and its mechanism.Methods The model of benign prostatic hyperplasia was established by subcutaneous injection of testosterone propionate 7 mg·kg^-1·d^-1 after bilateral testicular resection under anesthesia in mice.Male mice were randomly divided into six groups according to body weight:normal group,model group,control group(finasteride 1.4 mg·kg^-1·d^-1),and low,medium and high dose experimental groups(Pueraria isoflavones 50,100,200 mg·kg^-1·d^-1).On the second day of modeling,each groupwas given distilled water,finasteride and different doses of Pueraria isoflavones for 4 weeks.Enzyme-linked immunosorbent assay(ELISA)was used to detect the activity of serum serum 5-alpha reductase(5 AR),the contents of tumor necrosis factor-alpha(TNF-α)and interleukin-8(IL-8)in prostate tissue of mice.The contents of superoxide dismutase(SOD)and malondialdehyde(MDA)in serum of mice were determined by spectrophotometry.Results The activities of 5 AR in normal group,model group,control group and low,medium,high dose experimental groups were(2.46±0.06),(2.64±0.10),(2.51±0.08),(2.56±0.03),(2.48±0.05),(2.53±0.04)U·L^-1,respectively;the levels of TNF-αin the above six groups were(102.96±7.17),(118.56±5.16),(100.59±5.46),(104.11±2.68),(103.03±3.52),(102.54±9.95)pg·m L^-1,respectively;the levels of IL-8 in the above six groups were(9.07±0.54),(10.81±0.67),(9.46±0.74),(9.78±0.31),(9.28±0.55),(9.21±0.73)pg·m L^-1,respectively;the activity of SOD in the above six groups were(240.76±27.81),(173.03±14.48),(249.21±18.12),(218.55±31.20),(255.53±19.52),(219.06±16.90)U·m L^-1,respectively;the serum levels of MDA in the above six groups were(7.64±1.16),(11.58±0.68),(9.51±1.06),(9.23±1.12),(8.91±0.82),(9.90±1.50)nmol·m L^-1,respectively.Comparison between model group and normal group,the difference of the indicators were significantly(all P<0.01);comparison between the three dose experimental grou
