摘  要:目的:探讨中药解酒方葛根散对酒精性勃起功能障碍(ED)大鼠阴茎平滑肌组织结构一氧化氮合酶(NOS)活力及α-平滑肌肌动蛋白(α-SMA),缝隙连接蛋白基因43(Cx43),转化生长因子-β1(TGF-β1)m RNA和蛋白表达的影响,为临床应用葛根散治疗酒精性ED提供实验依据。方法:将SD大鼠随机分为5组,分别为正常组、模型组、葛根散低、中、高剂量组(5,10,20 g·kg^-1);除正常组外,其他各组按照15 m L·kg^-1·d^-1给予乙醇干预30 min后给药。比色法检测酒精性ED大鼠阴茎平滑肌组织NOS活力;实时荧光定量聚合酶链式反应法(Real-time PCR),蛋白免疫印迹法(Western blot)检测酒精性ED大鼠阴茎平滑肌组织α-SMA,Cx43,TGF-β1mRNA及蛋白表达的情况。结果:与正常组比较,模型组大鼠阴茎平滑肌组织NOS活力,α-SMA,Cx43 m RNA及蛋白表达量显著下降(P<0.05),TGF-β1m RNA及蛋白表达量显著上升(P<0.05)。与模型组比较,葛根散低、中、高剂量组均可显著上调酒精性ED大鼠阴茎组织结构NOS活力及Cx43 m RNA及蛋白表达量(P<0.05),同时可显著下调TGF-β1mRNA及蛋白表达量(P<0.05),葛根散中、高剂量组均可显著上调酒精性ED大鼠阴茎组织α-SMA m RNA及蛋白表达量(P<0.05)。结论:葛根散对酒精性ED大鼠阴茎平滑肌组织结构有明显的保护作用,其具体机制可能与调节NOS活力及α-SMA,Cx43,TGF-β1mRNA及蛋白表达量有关。Objective:To investigate the activity of nitric oxide synthase(NOS)andα-smooth actin(α-SMA)in rat penile smooth muscle tissue of rats with alcoholic erectile dysfunction(ED).The effects of protein gene 43(connexin43,Cx43)and transforming growth factorβ1(TGF-β1)m RNA and protein expressions provide an experimental basis for the clinical application of Gegensan in the treatment of alcoholic ED.Method:SD rats were randomly divided into five groups:normal group,model group,and low,medium,high-dose Gegensan groups(5,10,20 g·kg^-1).Except the normal group,the other groups were administered with drugs after alcohol intervention for 30 min at 15 m L·kg^-1·d^-1.Colorimetric assay was used to detect NOS activity in the penile smooth muscle tissue of alcoholic ED rats.Quantitative real-time fluorescence polymerase chain reaction(Real-time PCR)and Western blot were used to detectα-SMA,Cx43,TGF-β1 m RNA and protein expressions in smooth muscle tissue of alcoholic ED rats.Result:Compared with the normal group,the expressions of NOS,α-SMA and Cx43 m RNA and protein in the penile smooth muscle of the model group decreased significantly(P<0.05),while the expressions of TGF-β1 m RNA and protein increased significantly(P<0.05).Compared with the model group,low/medium/high-dose Gegensan groups could significantly up-regulate the NOS activity and Cx43 m RNA and protein expressions in the penile tissue of alcoholic ED rats(P<0.05),while significantly down-regulate TGF-β1 m RNA expression,andα-SMA m RNA and protein expressions in the penis tissue of rats with alcoholic ED were significantly up-regulated(P<0.05).Conclusion:Gegensan has an obvious protective effect on the structure of penile smooth muscle of alcoholic ED rats.The specific mechanism may be related to the regulation of NOS activity and a-SMA,Cx43 and TGF-β1 m RNA and protein expressions.
