优质鸡SESN1基因单核苷酸多态性及其与生长性状的关联分析

摘  要:为探究SESN1基因单核苷酸多态性与优质鸡生长性状的相关性,研究采用直接测序法扩增SESN1基因CDS区前2 000 bp调控区域,结果发现9个单核苷酸多态性位点。通过对这些位点进行连锁分析,划分为C-1365T^G-1543C、G-1177A^C-1294A、T-1058G^T-1076A组成的3个单倍型块。通过分析突变位点与温氏N301家系生长性状的相关性,发现位点C-1365T、T-1076A、G-1058T均与温氏N301家系胸肌重达显著相关(P<0.05)。结果表明,SESN1基因与家鸡肌肉生长密切相关,上述多态位点可作为研究肉鸡胸肌重指标的潜在分子标记。In order to study the correlation between the polymorphism of SESN1 gene and chicken growth traits,the 5′-flanking region of chicken SESN1 was amplified by direct sequencing, and a total of nine SNPs were identified.Three haplotype blocks consisting of C-1365 T^G-1543 C, G-1177 A^C-1294 A, and T-1058 G^T-1076 A, were divided by linkage analysis. Association analysis with growth traits found three SNPs(C-1365 T, T-1076 A and G-1058 T) were significantly associated with breast muscle weight(P<0.05) of Wenshi N301 chicken. The results suggested that SESN1 gene was highly associated with muscle growth, might play a crucial role in the growth and development of skeletal muscle.
