摘 要:目的:研究养阴益气合剂对肺间质纤维化大鼠血清转化生长因子-β1(TGF-β1)、结缔组织生长因子(CTGF)、白细胞介素(IL)-18表达影响。方法:将120只大鼠随机分为假手术组,模型组,泼尼松组,养阴益气合剂低、中、高剂量组,每组20只,采用气管滴注博来霉素的方法复制肺纤维化大鼠模型;造模后第2天予药物灌胃干预。分别于第14、28天分批处死大鼠,留取肺组织和血清,行HE染色和Masson染色,ELISA法检测TGF-β1、CTGF、IL-18蛋白表达。结果:与模型组比较,养阴益气合剂高剂量能够明显减轻大鼠肺泡炎和纤维化的程度(P<0.01);第28天时,养阴益气各剂量均能显著降低血清TGF-β1、CTGF的表达(P<0.01,P<0.05);但对IL-18表达影响不明显。结论:养阴益气合剂可能通过减少TGF-β1、CTGF表达来抑制肺纤维化进展。Objective: To investigate the effects of Yangyin Yiqi Mixture on transforming growth factor-beta1(TGF-β1), connective tissue growth factor(CTGF) and interleukin-18(IL-18) in serum of pulmonary fibrosis rats. Methods: A total of 120 rats were randomly divided into 6 groups: control group, model group, prednisone group, low-dose, mid-dose, and high-dose Yangyin Yiqi Mixture groups(n=20). Bleomycin was used to establish the pulmonary fibrosis model rats. From the second day after modeling, each group was given a corresponding drug gavage for 28 days. The rats were killed on the 14 th and 28 th day respectively. The lung tissues were collected to make HE staining and Masson staining, to observe the pathological changes. Serum was collected to detect the expression of TGF-β1, CTGF and IL-18 by ELISA method. Results: Compared with model group, high-dose Yangyin Yiqi Mixture can significantly relieve the degree of alveolitis and fibrosis in sera of rats with pulmonary fibrosis(P<0.01). On the 28 th day, Yangyin Yiqi Mixture can decrease the expression of TGF-β1 and CTGF(P<0.01, P<0.05). However, there was no significant effect on the expression of IL-18. Conclusion: Yangyin Yiqi Mixture may inhibit the progression of bleomycin-induced pulmonary fibrosis by reducing the expression of TGF-β1 and CTGF.
