摘 要:目的:观察遹肺饮及其拆方对博莱霉素致肺纤维化大鼠转化生长因子β1(TGF-β1)/Smads信号通路的影响。方法:SPF级雄性SD大鼠144只随机分为假手术组、模型组、遹肺饮组及遹肺饮3拆方组(补气通络组、养阴通络组、祛瘀通络组)。气管内滴注博莱霉素复制肺纤维化大鼠模型,并于术后次日灌胃。于实验第8、15、29天未给药时分别取肺组织,行HE染色,ELISA法检测TGF-β1含量,放射免疫法检测层粘连蛋白(LN)含量,免疫组织化学染色观察Smad2、Smad3、Smad4表达情况。结果:与假手术组同期比较,模型组大鼠LN与TGF-β1含量明显升高(P<0.01),Smad2、Smad3、Smad4平均光密度明显增加(P<0.01);与模型组同期比较,各用药组大鼠在各时期肺组织LN与TGF-β1含量均有不同程度下降(P<0.05,P<0.01),Smad2、Smad3、Smad4平均光密度均有不同程度减少(P<0.05,P<0.01),其中以遹肺饮组效果最佳,3拆方组以补气通络组作用最优。结论:遹肺饮可通过调控TGF-β1/Smads信号通路抑制肺纤维化病理进程。Objective:To observe the effects of Yuefei Decoction and its decomposed formulas on TGF-β1/Smads signaling pathway in rats with pulmonary fibrosis induced by bleomycin.Methods:A total of 144 SPF level SD rats were randomly divided into sham operation group,model group,Yuefei Decoction group and Yuefei Decoction three decomposed formula group(Buqi Tongluo Group,Yangyin Tongluo Group,Quyu Tongluo Group).Intratracheal instillation of bleomycin was conducted to replicate the rat model of pulmonary fibrosis,and the drugs were intragastrically the next day after surgery.Lung tissue was taken on the 8 th,15 th and 29 th day of the experiment.HE staining and ELISA were used to detect laminin(LN)and transforming growth factor beta 1(TGF-β1).Immunohistochemical staining was performed to observe the expression of Smad2/3/4.Results:In the model group,the content of LN and TGF-β1 increased significantly(P<0.01),and average integral absorbance of Smad2/3/4 elevated significantly(P<0.01)compared with sham operation group.In comparison with model group,the contents of LN and TGF-β1 of the treatment group were decreased(P<0.05,P<0.01),average integral absorbance of Smad2/3/4 descent(P<0.05,P<0.01).The effect of Yuefei Decoction is better than the other treatment groups,Yiqi Tongluo group is the best in the three decomposed formula group.Conclusion:Yuefei Decoction can inhibit the pathological process of pulmonary fibrosis by regulating the signaling pathway of TGF-β1/Smad2/3/4.
