摘  要:目的观察变应性鼻炎(AR)大鼠翼腭神经节和鼻黏膜中垂体腺苷酸环化酶激活多肽(PACAP)的表达变化,并探讨其意义。方法选择20只大鼠采用卵清蛋白致敏方法制作AR模型。然后将20只大鼠分为AR组及干扰组,干扰组于AR模型制作成功后第2天,以A型肉毒毒素(BTX-A)盐水浸润的棉条局部填塞双鼻腔7 d,1次/d,共7 d。对照组大鼠仅鼻腔滴入生理盐水21 d。观察大鼠行为学表现、并进行行为学评分,免疫组化检测各组大鼠鼻黏膜和翼腭神经节PACAP蛋白,HE染色观察鼻黏膜和翼腭神经节炎症反应情况,ELISA法检测血清炎症因子(IFN-γ、IL-4、IFN-γ/IL-4)。结果AR组大鼠典型行为评分高于干预组和对照组(P均<0.05)。AR组翼腭神经节和鼻黏膜中PACAP阳性细胞数高于对照组和干预组(P均<0.01);干预组鼻黏膜PACAP阳性细胞数高于对照组(P<0.01)。AR组大鼠鼻黏膜中纤毛倒伏缺失、炎性细胞浸润、腺腔内炎性细胞渗出较干预组和对照组严重(P均<0.05)。AR组血清IFN-γ、IL-4水平及IFN-γ/IL-4较干预组和对照组高(P<0.01或0.05)。结论AR大鼠翼腭神经节及鼻黏膜组织PACAP表达均升高,其原因可能是翼腭神经节内的PACAP释放于鼻黏膜、且刺激炎性因子表达而参与AR的发生。Objective To observe the expression changes of pituitary adenylate cyclase-activating polypeptide(PACAP)in the pterygopalatine ganglion and nasal mucosa in allergic rhinitis(AR)rats and to explore its significance.Methods Thirty Sprague-Dawley rats were randomly divided into 3 groups:the AR group,the intervention group,and the control group,with 10 in each.In the AR group,ovalbumin was used to sensitize the healthy rats.In the intervention group,botulinum toxin type A(BTX-A)was dripped into the nasal cavity of AR rats.In the control group,only physiological saline was used to drip into the nasal cavity of AR rats.Changes of the rats’behavior were observed.Histopathology and immunohistochemistry were employed to observe morphology in the rats’nasal mucosal and pterygopalatine ganglion,as well as the expression of PACAP.Results The typical behavioral scores of the AR group were higher than those of the intervention group and the control group(P<0.05).The PACAP positive cells in the rats’pterygopalatine ganglion and nasal mucosa of the AR group were more than those of the intervention group and control group(all P<0.01),and the PACAP positive cells of the AR group were more than those of the control group(all P<0.01).The cilium loss,inflammatory cells infiltration,and inflammatory cells exudation of nasal mucosa were severer in the in the AR group than in the intervention group and control group(all P<0.01).Conclusion The expression of PACAP increases in the pterygopalatine ganglia and nasal mucosa of AR rats,because PACAP in the pterygopalatine ganglion is released into the nasal mucosa and stimulates the expression of inflammatory factors and participates in the occurrence of AR.
