摘 要:目的通过观察生物卵膜对小鼠皮肤缺损创面的修复作用,来探讨该生物卵膜在耳科乳突术腔中应用的可能性。方法将100只健康雄性C57BL/6小鼠随机分为2mm组和3mm组,各50只,腹腔麻醉,在背部制备3个深及筋膜的直径2mm或3mm的圆形皮肤缺损创面,将其随机分为5组,生物卵膜组,生物卵膜+蛋清液组,莫匹罗星(商品名:百多邦)组,聚维碘酮(碘伏)组和对照组。对照组不予生物卵膜覆盖且不涂抹药物。对比相同大小皮肤缺损的愈合情况。于造模后12天后取2mm组小鼠皮损部位,予4%多聚甲醛固定液充分固定,进行石蜡包埋切片,观察各组皮肤创面愈合的病理结构。结果随着时间的延长,五组残余创面面积逐渐减小,在2mm组,生物卵膜组和聚维碘酮组创面愈合率高于其余三组,生物卵膜组和聚维碘酮组的愈合时间短于其余组。在3mm组,生物卵膜组的创面愈合率高于包括聚维碘酮组在内的其余四组,生物卵膜组的愈合时间最短,差异有统计学意义(P<0.05)。生物卵膜组组织病理学变化较对照组皮肤有较大改善,接近正常皮肤组织形态。结论生物卵膜可以明显促进创面愈合速度,为临床皮肤损伤修复提供新的治疗方法,存在在耳科乳突术腔修复应用的潜能。Objective To investigate the potential of biological egg membrane in skin defect repair and possible application in the mastoid surgery cavity using a mouse model. Methods One hundred healthy male C57 BL/6 mice were randomly assigned to a 2 mm and a 3 mm diameter skin wound group(n=50 in each group). The circular skin wound reached the fascia on the back under general anesthesia. Animals in each group were randomly divided into 5 groups based on wound treatment, i.e. biological egg membrane, biological egg membrane + egg white solution, Mupirocin, iodophor and no treatment(control). Healing of the skin defect of the same size was compared. After 12 days, the skin lesion in mice in the 2 mm skin wound group was taken, fixed in 4% paraformaldehyde, embedded in paraffin, and sectioned for structural examination. Results Along with time, the area of skin defect decreased gradually under all five treatment conditions. In the 2 mm skin wound group, healing was faster in biological egg membrane and iodophor treated animals than in those treated otherwise, resulting in shorter healing time. In the 3 mm skin wound group, healing was fastest in the biological egg membrane treated animals, faster than even those treated with iodophor group, with the shortest healing time. Histopathological examination showed significantly improved healing in biological egg membrane treated animals compared with those in the control group(P<0.05), approaching normal skin tissue morphology.Conclusion Biological egg membrane can promote skin wound healing and provide a new therapeutic approach to skin defect repair and potentially repair of mastoid cavities.
