摘 要:[目的]以厚朴/雷公藤(处理①)、杉木/雷公藤(处理②)、马尾松/雷公藤(处理③)为3种套种模式,雷公藤纯林为CK对照组,建立不同套种模式下雷公藤甲素的HPLC测定方法,并对不同套种模式下雷公藤不同部位甲素的含量进行比较。[方法]采用高效液相色谱法(HPLC),色谱柱为SinoChrom ODS-BP(4.6 mm×250 mm,5μm);流动相为甲醇-水(体积比45∶55);流速1.0 mL/min;检测波长218 nm;柱温40℃;进样量20μL。[结果]该方法能在8 min左右实现雷公藤甲素的快速分离,雷公藤甲素浓度在1.55~248.00μg/mL与峰面积线性关系良好(R 2=0.9997),平均加样回收率为99.6%,RSD为1.85%(n=6)。[结论]该方法易操作、分析速度快、重现性好、准确率高,可作为雷公藤甲素含量的测定方法。不同套种模式下雷公藤甲素总含量从高到低依次为处理①、CK、处理②、处理③,雷公藤不同部位的雷公藤甲素含量从高到低依次为根、叶、茎,说明厚朴林下套种雷公藤的模式可提高雷公藤甲素的含量,因此可作为优质林药间作模式广泛推广。[Objective]There were three interplanting patterns of Magnolia officinalis/Tripterygium wilfordii(Treatment 1),Cunninghamia lanceolata/Tripterygium wilfordii(Treatment 2),Pinus massoniana/Tripterygium wilfordii(Treatment 3),and the pure forest of Tripterygium wilfordii was CK control group.HPLC method for the determination of triptolide in different interplanting patterns was established,and compared triptolide content in different parts of Tripterygium wilfordii.[Method]High performance liquid chromatography method was used,a SinoChrom ODS-BP 5μm column(4.6 mm×250 mm)was used at 40℃;mobile phase was methanol-water(volume ratio 45:55);the flow rate was 1.0 mL/min;the detection wavelength was 218 nm;the injection volume was 20μL.[Result]The method could achieve rapid separation of triptolide in about 8 minutes.A good linear was obtained within the range of 1.55-248.00μg/mL(R 2=0.9997),the average recovery was 99.6%(RSD=1.85%,n=6).[Conclusion]The method is easy to operate,has fast analysis speed,good reproducibility and high accuracy,and can be used as a method for determining the content of triptolide.The order of total content of triptolide in different interplanting patterns is treatment 1>CK>treatment 2>treatment 3,and the order of triptolide in different parts of Tripterygium wilfordii is root>leaf>stem.It shows that the model of Tripterygium wilfordii under Magnolia officinalis can increase the content of triptolide,so it can be widely promoted as a high quality forest medicine interplanting pattern.
