摘　　要：本试验主要探讨了鹿茸间充质干细胞条件培养基对乳腺癌细胞4T1的作用。分离、培养、鉴定了鹿茸间充质干细胞,采用MTT法检测了鹿茸间充质干细胞条件培养基对4T1细胞的增殖活性的影响,并通过Western blot检测了对4T1细胞凋亡的影响。结果显示,鹿茸间充质干细胞条件培养基可抑制4T1细胞的增殖,并呈浓度依赖性,并伴有4T1细胞内Cleaved-caspase 3蛋白表达上调以及Bcl-xl蛋白表达的下调。这一结果表明,鹿茸间充质干细胞条件培养基可抑制4T1细胞增殖,其机制与细胞内凋亡信号通路有关。This study mainly investigated the effect of antler mesenchymal stem cells conditioned medium on the breast cancer cell line 4T1.Antler mesenchymal stem cells were isolated,cultured and identified,the effect of the conditioned medium of antler mesenchymal stem cells on the proliferation activity of 4T1 cells was detected by MTT,the apoptosis of 4T1 cells was detected by Western blot.The results showed that antler mesenchymal stem cells conditioned medium could inhibit the proliferation of 4T1 cells in a dose-dependent manner,and these effects were associated with up-regulation of cleaved-caspase 3 expression and down-regulation of Bcl-xl expression in 4T1 cells.These results suggest that the antler mesenchymal stem cells conditional medium can inhibit the proliferation of 4T1 cells,which related to the intracellular apoptosis signaling pathway.
