摘 要:检测阿片黑皮质素前体基因部分外显子区单核苷酸多态性(Single nucleotide polymorphisms,SNPs)在不同水貂品种中的分布,探讨该基因SNPs与水貂生长性状的相关性.以咖啡水貂和红眼白水貂共计179个样本的血液基因组DNA为模板,采用PCR扩增及直接测序技术对POMC基因SNPs进行检测,并将变异位点与水貂体重、体长进行关联分析.引物P1扩增片段存在g.171A>G、g.470T>G 2个SNPs;引物P2扩增片段存在g.514C>T 1个SNPs.3个SNPs在2个水貂品种中表现为中度多态(0.25<PIC<0.50),且均处于Hardy-Weinberg平衡状态.关联分析表明,g.171A>G位点与咖啡水貂体重显著相关(P<0.05);g.470T>G位点与咖啡水貂体长显著相关(P<0.05),与红眼白水貂体重显著相关(P<0.05);g.514C>T位点与红眼白水貂体长显著相关(P<0.05).POMC基因可能是影响水貂生长性状的关键基因.To detect the polymorphism of POMC gene in different breeds mink and analyze the relationship between the genetic polymorphisms and growth traits of mink,the blood samples of 179 mink from either coffee color in hair or pink eyed as well as white hair mink were collected and their genomic DNA were extracted.The SNPs were detected using PCR amplification and sequencing techniques.Furthermore,the relationship between the loci and body weight(length)were analyzed.g.171 A>G and g.470 T>G mutation sites were found in primes 1 amplified fragments.g.514 C>T mutation site was found in primes 2 amplified fragments.The three SNPs were at moderate polymorphic status in coffee mink and pink eyed white mink.The association of polymorphisms revealed that g.171 A>G loci had significant correlation with body weight of coffee mink,g.470 T>G loci had significant correlation with body length of coffee mink,g.470 T>G loci had significant correlation with body weight of pink eyed white mink,g.514 C>T loci had significant correlation with body length of pink eyed white mink.POMC gene maybe a key gene affecting mink growth traits.
