基于网络药理学研究黄连治疗心房颤动的活性成分及作用机制

摘　　要：目的探究黄连治疗心房颤动的药效物质基础、作用机制。方法检索黄连的化学成分及作用靶点及心房颤动的靶基因,构建“成分-靶点-疾病网络”。进而行GO功能富集分析及KEGG通路富集分析,分析黄连治疗心房颤动的活性成分、作用靶点及机制。结果黄连的11个活性成分作用于心房颤动的159个靶点,涉及20条信号通路。黄连活性成分靶点可能通过基因表达、蛋白代谢、巨核细胞分化等生物过程,调节PI3K-Akt、丝裂原活化蛋白激酶(MAPK)、低氧诱导因子-1(HIF-1)、Notch等信号通路来发挥治疗心房颤动的作用。结论初步揭示黄连治疗心房颤动的有效成分和作用靶点,为开发更好的心房颤动治疗候选药物提供参考依据。Objective To explore the pharmacodynamic material basis and mechanism of Coptis chinensis in the treatment of atrial fibrillation.Methods The chemical constituents of Coptis chinensis and target genes of Coptis chinensis and atrial fibrillation were retrieved,and the"Component-Target-Disease Network"was constructed.Then GO function enrichment analysis and KEGG pathway enrichment analysis were performed to analyze the active ingredients,targets and mechanisms of Coptis chinensis in the treatment of atrial fibrillation.Results There were 11 active components of Coptis chinensis acted on 159 targets of atrial fibrillation and involved 20 disease signaling pathways.The target of Coptis chinensis active ingredients might play a role in the treatment of atrial fibrillation by regulating PI3K-AKT,MAPK,HIF-1,Notch and other signaling pathways through gene expression,protein metabolism,megakaryocyte differentiation and other biological processes.Conclusion The active components and targets of Coptis chinensis against atrial fibrillation were preliminarily revealed,which provided reference for the development of better atrial fibrillation candidates drugs.
