摘 要:目的:研究黄芪多糖(astragaluspolysacharin,APS)对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC)周期及血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法:用不同浓度黄芪多糖对HUVEC进行干预培养,流式细胞仪检测细胞周期以及细胞凋亡;荧光倒置显微镜检测细胞表达VEGF的免疫荧光。结果:MTT法表明,APS在一定的范围内(0.1~100μg/mL)能以剂量依赖方式促进HUVEC细胞周期从G0/G1期向G2/M期和S期转变,流式细胞凋亡检测显示APS并未增加HUVEC凋亡率。细胞免疫荧光染色实验结果表明,APS促进VEGF表达。结论:APS在一定的浓度范围内(0.1~100μg/mL)促进HUVEC细胞分裂增殖。APS还能上调HUVEC细胞VEGF的表达水平,为进一步血管化提供有效的促血管化因子,为治疗缺血性疾病提供新思路。Objective To explore the effect of Astragalus polysaccharide(APS)on the cell cycle of human umbilical vein endothelial cells(HUVEC)and the expression of vascular endothelial growth factor(VEGF).Methods HUVEC were cultured with different concentration of APS,and the cell cycle and apoptosis were detected by flow cytometry.Immunofluorescence staining of VEGF in cells was detected by fluorescence microscope.Results MTT assay showed that APS promoted the cell cycle of HUVEC from G0/G1 phase to G2/M phase and S phase within a certain concentration range.Flow cytometry apoptosis assay showed that APS did not increase the apoptosis rate of HUVEC.Cellular immunofluorescence staining showed that APS promoted VEGF expression.Conclusion APS could promote division and proliferation of HUVEC at a certain concentration range(0.1–100μg/mL).In addition,APS can also up-regulate the expression level of VEGF in HUVEC,providing an effective angiogenic factor for further vascularization and a new idea for the treatment of ischemic diseases.
