摘 要:目的研究土荆皮中具有抗黄嘌呤氧化酶活性的化学成分。方法采用硅胶、Sephadex LH-20、反相柱色谱(ODS)、高效液相色谱(HPLC)制备等方法对土荆皮的乙醇提取物进行分离纯化,根据化合物的波谱[核磁共振氢谱(1H-NMR),核磁共振碳谱(13C-NMR),高分辨电喷雾电离质谱(HRESI-MS)]数据进行结构鉴定,采用HPLC测定化合物的黄嘌呤氧化酶抑制活性。结果从土荆皮乙醇提取物的乙酸乙酯萃取部位分离得到13个化合物,分别鉴定为土荆皮乙酸(1),土荆皮甲酸(2),土荆皮丙酸(3),土荆皮乙酸甲酯(4),土荆皮乙酸乙酯(5),土荆皮乙酸葡糖苷(6),杨梅素(7),二氢去氢二愈创木基醇(8),罗汉松脂素(9),cedrusin(10),ligraminol E(11),原儿茶酸(12),3,5-二羟基-2-[2-(4-羟苯乙酰基)]苯甲酸(13)。化合物7,12,13的黄嘌呤氧化酶抑制活性较好,半数抑制浓度(IC50)分别为0.32,0.38,0.29 mmol/L。结论化合物8,9,11,12,13为首次从土荆皮中分离得到,其所含的多酚类化合物具有较好的抗黄嘌呤氧化酶活性。Objective To investigate the chemical constituents from Pseudolaricis cortex xanthine oxidase(XO)inhibitory activity.Methods The compounds were isolated and purifide by silica gel,Sephadex LH-20,ODS column chromatography and preparative HPLC.Their structural identification was performed on the basis of their 1H-,13C-NMR and HRESI-MS data.In vitro XO inhibitory activity was detected by an HPLC method.Results Thirteen constituents in the ethyl acetate extract were isolated and their structures were identified as pseudolaric acid B(1),pseudolaric acid A(2),pseudolaric acid C1(3),pseudolaric acid B methyl ester(4),pseudolaric acid B ethyl ester(5),pseudolaric acid B-O-β-D-glucopyranoside(6),myricetin(7),dihydrodehydrodiconiferyl alcohol(8),matairesinol(9),cedrusin(10),ligraminol E(11),protocatechuic acid(12),3,5-dihydroxy-2-[2-(4-hydroxy-phenyl)acetyl]benzoic acid(13).Compounds 7,12 and 13 revealed the potential XO inhibitory activity with IC50 values at 0.32,0.38 and 0.29 mmol/L,respectively.Conclusion Compounds 8,9,11,12,13 were obtained from Pseudolaricis cortex for the first time and polyphenols in Pseudolaricis cortex might be responsible for the XO inhibitory activity.
