摘 要:目的:采用高效液相色谱法对民族药材大菟丝子中绿原酸含量进行测定并开展方法学研究;方法:以乙腈-0.05%磷酸溶液(10∶90)为流动相,等度洗脱,流速1.0 mL/min,检测波长327nm,采用C18色谱柱,柱温30℃,外标法定量。结果:绿原酸在0.5275~105.5μg/mL浓度范围内与峰面积呈良好的线性关系(r=0.9994),平均回收率为99.2%(RSD=0.5%)。结论:测定方法快速、简便,符合方法验证要求,可用于大菟丝子中绿原酸含量的测定和质量控制。Objective Establish a method for determination of chlorogenic acid in Cuscuta Japonica Choisy with HPLC. Methods The mobile phase was consisted of acetonitrile-0.05% phosphoric acid aqueous solution(10∶90), The flow rate was 1.0 mL/min, the detective wavelength was 327 nm and the C18 column temperature was 30℃.With external standard method to make quantitative analysis.Results The linear range of the method was 0.5275 ~105.5 μg/mL, the average recovery was 99.2%,RSD was 0.5%. Conclusion This method is simple, fast and efficient, which will be used to provide accurate, scientific quality control for determination of chlorogenic acid in Cuscuta Japonica Choisy.
