摘  要:目的长期大量饮酒对小鼠麻醉敏感性的影响以及其相关机制的探讨。方法采用随机分组对照研究的方法,将30只雄性C57小鼠随机分为对照组和实验组。对照组给予正常饮食,实验组将用5%酒精(v/v)持续自主饮酒+3.5g·kg^-1·3天灌胃(20%酒精(v/v))代替正常饮水,其余处理与对照组相同。30天后,比较对照组和实验组丙泊酚和异氟烷的麻醉敏感性差异,同时检测线粒体ComplexⅡ的功能,探讨长期大量饮酒对小鼠麻醉敏感性以及线粒体功能的影响。结果与对照组比较,实验组小鼠对丙泊酚和异氟烷的敏感性均降低(丙泊酚ED50:7.71mg/kg vs.9.34mg/kg,异氟烷EC50:0.79%vs.0.92%),实验组线粒体耗氧率(OCR,oxygen consumption rates)(%)值明显降低,差异有统计学意义[(233.3±19.17)vs.(160.2±7.46),P<0.05]。结论长期大量饮酒通过影响线粒体complexⅡ氧化磷酸化过程,导致ATP生成减少,影响线粒体功能,导致丙泊酚和异氟烷的麻醉敏感性降低。Objective To investigate the effects of long-term heavy drinking on anesthesia sensitivity in mice and its related mechanisms.Methods Thirty male C57 mice were randomly divided into control group(CG)and experimental group(EG)based on the methods of randomized controlled study.CG was given a normal diet,and EG was treated with 5%alcohol(v/v)for continuous drinking+3.5 g·kg^-1·3 days of gavage(20%alcohol(v/v))instead of normal drinking water.Other treatments were the same as CG.After 30 days later,the anesthesia sensitivity of propofol and isoflurane were compared,and mitochondrial ComplexⅡfunction was also examined,which aimed to investigate the effects of long-term heavy drinking on anesthesia sensitivity and mitochondrial function in mice.Results Compared with CG,the mice in EG were less sensitive to propofol and isoflurane(Propofol ED50:7.71 mg/kg vs.9.34 mg/kg,Isoflurane EC50:0.79%vs.0.92%),the mitochondrial oxygen consumption rate(OCR)value of EG was significantly lower[(233.3±19.17)vs.(160.2±7.46),P<0.05].Conclusion Long-term heavy drinking could lead to reduce ATP production,affect mitochondrial function and reduce anesthesia sensitivity of propofol and isoflurane through affecting the oxidative phosphorylation process of mitochondrial complexⅡ.
