摘  要:目的肿瘤细胞的耐药性严重限制了化疗药物的临床应用,本研究成果表明,将化疗药物“敏感剂”与化疗药物联合应用可以有效地增加化疗药物的抗肿瘤活性。方法构建同时负载阿霉素(DOX)与化疗药物“敏感剂”姜黄素(Cur)的纳米递药系统,采用Zeta电位及纳米粒度分析仪、紫外-可见分光光度计及激光共聚焦显微镜等对纳米粒的形态、体外释药、体外抗肿瘤活性及纳米粒转运DOX在肿瘤细胞的分布等方面进行考察和评价。结果制得纳米粒的粒径为(188±12)nm,DOX与Cur的载药量分别为5%和10%,纳米粒的体外药物释放呈现pH依赖性,细胞实验结果显示Cur可以有效地增加DOX在耐药肿瘤细胞内的分布,因此显示出更高的细胞毒性。结论化疗药物“敏感剂”与化疗药物的联合应用对耐药肿瘤细胞有较好的疗效,具有进一步研究的价值。Objective Drug resistance of tumor cells severely limits the clinical use of chemo-therapeutic drugs.A combinational therapy consisting of chemo-sensitizer drugs and chemotherapy drugs could increase the antitumor activity of chemotherapeutic agents.Methods In this study,we developed doxorubicin(DOX)and curcumin(Cur)co-delivery nanoparticle,then used the zeta potential and particle size analyzer,UV-visible spectrophotometer and laser confocal microscope to investigate and evaluate the morphology of the nanoparticles in vitro drug release and in vitro anti-tumor active transport of DOX of tumor cells.Results We successfully made the nanoparticle with the average size of(188±12)nm and the drug loading of DOX and Cur 5%and 10%.Drug release of nanoparticles in vitro is pH-dependent.The cell experimental results show that Cur can increase the distribution of DOX in drug resistant tumor cells,thus showing higher cytotoxicity.Conclusion The combination of chemo-sensitizer drugs and chemotherapy drugs has certain curative effect on drug-resistant tumor cells,and has further research value.
