摘  要:目的探讨中药复方凉血固元汤(LGD)对肠道急性辐射损伤大鼠肠道菌群的调节作用。方法选择81只雌性SD大鼠,随机选取6只作为空白(control)组,其余大鼠采用60Co-γ射线照射制作肠辐射损伤模型,并随机分为模型(model)组、金双歧(GB)组及LGD组,每组25只。照射后每日观察大鼠排便、死亡等一般情况,于照射后7 d称取大鼠体质量,并收集粪便样本用于肠道菌群检测,分析各组肠道菌群的α多样性和门、属水平上主要菌群相对丰度差异。结果照射后3 d开始各照射组大鼠出现腹泻、死亡等情况,以模型组最明显,照射后7 d各照射组体质量显著低于空白组(P<0.01),LGD组高于模型组(P<0.05)。肠道菌群α多样性比较,模型组及GB组Shannon指数较空白组升高(P<0.05),Simpson指数降低(P<0.05,P<0.01),而LGD组Shannon指数低于模型组(P<0.01),Simpson指数较GB组升高(P<0.05)。门水平各组菌群丰度比较可见,与空白组比较,模型组及GB组厚壁菌门相对丰度明显降低(P<0.01),拟杆菌门显著升高(P<0.01),两者比例倒置,而LGD组厚壁菌门较模型组及GB组明显升高(P<0.01),且拟杆菌门显著降低(P<0.01)。在属水平上,模型组和GB组拟杆菌属相对丰度较空白组升高(P<0.01,P<0.05),LGD组低于模型组(P<0.01),且布劳特菌属较模型组升高(P<0.01)。结论LGD能有效调节辐射导致的肠道菌群的结构失调和数量变化,改善大鼠生存质量。Objective To investigate the regulatory effect of Liangxue Guyuan decoction(LGD)on intestinal flora in rats with acute intestinal radiation injury.Methods Totally 81 female SD rats were randomly selected,with 6 rats in the control group.The remaining rats were irradiated by 60Co-gamma rays to establish an intestinal radiation injury model.After that,they were randomly divided into the model group,Golden Bifid(GB)group and LGD group,with 25 rats per group.The defecation and death of rats were observed daily.On the 7th day after irradiation,the reacive body mass was measured and fecal samples were collected for intestinal flora detection.The alpha diversity of intestinal flora and the relative abundance of major flora at Phylum and Genus levels were analyzed.Results Diarrhea and death occurred in rats of each irradiation group from the 3rd day after irradiation,especially in the model group.The body mass of each irradiation group was significantly lower than that of the control group(P<0.01),and that of the LGD group was higher than that of the model group(P<0.05).Shannon index of model group and GB group was higher than that of control group(P<0.05),but Simpson index was lower(P<0.05,P<0.01).Shannon index of LGD was lower than that of model group(P<0.01),but Simpson index was higher than that of GB group(P<0.05).Compared with the control group,the relative abundance of Firmicutes in the model group and GB group was significantly lower(all P<0.01),Bacteroidetes were significantly higher(P<0.01),F/B ratio was inverted,while the proportion of Firmicutes in the LGD group was significantly higher(P<0.01)and Bacteroidetes were significantly lower(P<0.01).At the Genus level,the relative abundance of Bacteroides in the model group and the GB group was increased compared with the control group(P<0.01,P<0.05),that of the LGD group was lower(P<0.01),while Blautia in the LGD group increased(P<0.01).Conclusion Liangxue Guyuan decoction can effectively regulate the structural imbalance and quantity change in intestinal flora ca
