摘 要:为了探讨精神依赖物海洛因对胚胎大鼠形态结构发育和小脑c-Fos表达的影响,将受孕后的Wistar大鼠40只,随机分为对照组、海洛因低、中、高剂量组,每组10只孕鼠。于第7天时,分别给予32.5、65和130 mg·kg^-1的海洛因,连续9 d,观察该物对胚胎大鼠形态结构发育的影响,用免疫组化法检测胚胎大鼠小脑组织c-Fos基因表达水平。结果发现,海洛因低、中剂量组活胚胎总数比对照组分别减少15.87%和29.37%;海洛因低、中剂量组出现了多指,脐疝,双耳、肋骨、腰椎骨、前肢掌骨、脑发育不全等畸形胚胎;高剂量组孕鼠未见成型的胚胎。海洛因低、中剂量组胚胎小脑皮层中c-Fos表达水平(IOD)比对照组分别增加了48.94%和95.74%(P<0.01)。结果表明,精神依赖物海洛因能通过母体干扰胚胎表型和组织器官的发育,诱导胚胎小脑组织细胞c-Fos基因表达,损伤脑组织细胞的发育。To investigate the effects of spiritual dependence(heroin)on morphological structure development and c-Fos expression in cerebellar cortex of embryonic rats,the pregnant Wistar rats were randomly divided into control group and heroin drug group(divided into low,medium and high dose heroin groups).On the 7th day after conception,Wistar rats were treated with32.5,65 and 130 mg·kg^-1 heroin for 9 d.The effects of spiritual dependence(heroin)on the morphological and structural development of embryonic rats were observed.The c-Fos expression level in cerebellar cortex of embryonic rats was determined by immunohistochemical staining.The results showed that low,medium dose heroin groups,the total number of live embryos decreased by 15.87%and 29.37%respectively,compared with the control group.Multiple fingers,umbilical hernia,two ears,ribs,lumbar vertebrae,and forelimb metacarpal bone dysplasia occurred in low and medium dose groups of heroin;in high dose group of heroin no embryo was formed in pregnant rats.Cell imaging and immunohistochemical tests showed that c-Fos expression level(IOD)was increased by 48.94%and 95.74%,respectively,in the cerebellar cortex of low and middle dose groups of heroin,compared with the control group(P<0.01).The results showed that spiritual dependence(heroin)inhibits the morphological development and cerebellar c-Fos expression level in embryonic rats,and the inhibition increases with the increase of the dose to heroin.
