摘 要:脆性X智力低下1号基因(fragile X mental retardation 1,fmr1)编码的蛋白FMRP是一种RNA结合蛋白,当fmr1基因缺失时雌鼠卵巢中卵泡数目减少、生殖功能会降低。牙鲆(Paralichthys olivaceus)是中国重要的海水经济鱼类,其性腺发育一直颇受关注。为了探讨fmr1基因在牙鲆性腺发育中的作用,本研究运用生物信息学方法分析了fmr1基因的分子结构与进化特征,并采用荧光定量PCR技术检测了其在牙鲆发育中的表达情况,探讨了高温对牙鲆性腺分化中fmr1基因表达的影响。结果表明,牙鲆fmr1基因含有15个外显子和14个内含子,存在两个高度保守的KH(hnRNP K homology)结构域,该结构域是RNA结合蛋白的特征结构域。共线性及系统进化分析表明fmr1在鱼类中进化关系相对保守。荧光定量PCR结果显示fmr1在牙鲆成鱼雌雄性腺中表达量相对较高,且在卵巢中表达量最高;在不同发育时期中,fmr1在神经胚期表达量最高;且高温处理对fmr1在性腺分化早期的表达有一定的抑制作用。这些结果暗示了fmr1基因可能在牙鲆性腺发育中发挥着重要的作用。The protein FMRP encoded by fragile X mental retardation 1 protein FMRP is a RNA binding protein.The number of follicles ovaries and reproductive function decreased when the fmr1 gene was absent in the female mice.Paralichthys olivaceus is an important marine economic fish and the research on gonadal development has attracted wide attention.In order to explore the role of fmr1 gene in gonad development of P.olivaceus,we analyzed the molecular structure and evolution characteristics of fmr1 by bioinformatics,and then detected the gene expression of fmr1 during development of P.olivaceus using Real-time fluorescence quantitative PCR.Finally,we discussed the effect of high temperature on the gene expression of fmr1 during early gonad differentiation.The results showed that fmr1 gene of P.olivaceus contains 15 exons and 14 introns,and there are two highly conserved KH(hnRNP K homology)domains which is the characteristic domain of the RNA binding protein.Synteny and phylogenetic analyses indicated that the evolutionary relationship of fmr1 was relatively conservative in fish.Real-time fluorescence quantitative PCR results demonstrated that the expression of fmr1 gene is relatively higher in adult gonad,and it has significant high expression in ovary.In different stages of development, the highest expression of fmr1 gene was observed in neural stage. After the treatment of hightemperature, the expression of fmr1 gene were decreased in the early stage of gonad differentiation. The studyshowed that the fmr1 gene may play an important role in gonad development of P. olivaceus.
