摘 要:雷公藤内酯醇是从中草药雷公藤(Tripterygium wilfordii)中提取的环氧二萜内酯化合物,具有较强的抗癌作用,但在口腔癌的治疗中应用较少。本研究旨在探讨雷公藤内酯醇对口腔癌细胞增殖、侵袭、迁移和血管生成的影响。本研究用20μmol/L的雷公藤内酯醇预处理人舌鳞状细胞癌SAS细胞系6 h,然后分别进行生长抑制试验、侵袭试验和伤口愈合试验,并采用RT-qPCR检测雷公藤内酯醇预处理对血管内皮生长因子(VEGF)、白细胞介素-6(IL-6)、E-钙粘蛋白(E-cadherin)和基质金属蛋白酶-9(MMP-9)mRNA表达的影响。研究显示,在用不同浓度(0~100μmol/L)的雷公藤内酯醇处理SAS细胞12 h、24 h和48 h后,SAS细胞增值率随着雷公藤内酯醇浓度的升高而逐渐降低。与对照组相比,雷公藤内酯醇处理可导致SAS细胞侵袭率显著降低58.01%。雷公藤内酯醇处理可导致SAS细胞伤口面积显著降低64.04%。与对照组相比,雷公藤内酯醇处理组的VEGF mRNA表达量显著降低了52.12%。与对照组相比,雷公藤内酯醇处理组的IL-6 mRNA表达量显著降低了53.89%,E-cadherin mRNA表达量显著升高了331.28%,MMP-9 mRNA表达量显著降低了84.26%。本研究表明,雷公藤内酯醇对人舌鳞状细胞癌具有较好的抑制作用,有望成为口腔癌治疗的候选药物。Triptolide is an diterpenoid triepoxide compound extracted from the Chinese herbal medicine Tripterygium wilfordii,which has a strong anticancer effect,but it is less used in the treatment of oral cancer.This study was to investigate the effects of triptolide on proliferation,invasion,migration and angiogenesis of oral cancer cells.In this study,human tongue squamous cell carcinoma SAS cell line was pretreated with 20μmol/L triptolide for 6 h,RT-qPCR was used to detect the effects of triptolide pretreatment on the expression of vascular endothelial growth factor(VEGF),interleukin-6(IL-6),E-cadherin and matrix metalloproteinase-9(MMP-9).Studies have shown that after treatment of SAS cells with different concentrations(0~100μmol/L)of triptolide for 12 h,24 h and 48 h,the SAS cell proliferation rate decreases with the increase of triptolide concentration.Compared with the control group,triptolide treatment resulted in a significant reduction in SAS cell invasion rate of 58.01%.Treatment with triptolide resulted in a significant reduction in wound area of SAS cells of 64.04%.Compared with the control group,the expression of VEGF mRNA in the triptolide treatment group was significantly reduced by 52.12%.Compared with the control group,the expression of IL-6 mRNA in the triptolide treatment group was significantly decreased by 53.89%,the expression of E-cadherin mRNA was significantly increased by 331.28%,and the expression of MMP-9 mRNA was significantly decreased by 84.26%.This study shows that triptolide has a good inhibitory effect on human tongue squamous cell carcinoma,and it is expected to be a candidate for oral cancer treatment.
