摘 要:目的:探讨小檗碱抑制体外培养翼状胬肉成纤维细胞迁移的药理机制。方法:通过培养临床获取的翼状胬肉组织获得翼状胬肉成纤维细胞,采用终浓度0、20、40、80μmol/L的小檗碱对细胞进行处理,检测细胞迁移相关因子及AMP依赖的蛋白激酶(AMPK)途径关键因子(MMP-1、MMP-2、MMP-9、α-SMA、AMPK、ACC)mRNA及蛋白表达水平变化。结果:随着小檗碱终浓度的提高,AMPK、ACC mRNA表达水平及蛋白表达水平上调;细胞迁移能力降低;MMP-1、MMP-2、MMP-9及α-SMA等因子mRNA表达水平及蛋白表达水平显著降低。结论:小檗碱可通过激活AMPK信号通路抑制体外培养翼状胬肉细胞的迁移能力。Objective:To explore the pharmacological mechanism of berberine inhibiting the migration of in vitro pterygium fibroblasts.Methods:Pterygium fibroblasts were obtained from cultured clinically acquired pterygium tissues.Berberine with a final concentration of 0μmol/L,20μmol/L,40μmol/L,80μmol/L was used to treat the cells,and the mRNA and protein expression levels of cell migration related factors and AMPK key factors were examined.Results:The expressions of AMPK and ACC mRNA and protein were up-regulated and the expression levels of MMP-1,MMP-2,MMP-9 andα-SMA mRNA and protein were significantly decreased,with the increase of the final concentration of berberine.Conclusion:Berberine inhibits the migration of pterygium cells in vitro by activating AMPK signaling pathway.
